Summary: This study presents the design of tetraspecific molecules that can activate natural killer cells for cancer therapy. The tetraspecific molecules selectively activate and proliferate natural killer cells, and the binding to tumor-associated antigens triggers cell cytotoxicity and cytokine production. In animal experiments, the tetraspecific molecules control tumor growth and accumulate at the tumor site. Furthermore, they can deplete circulating B cells without causing toxicity in non-human primates.

IL-17-dependent fibroblastic reticular cell training boosts tissue protective mucosal immunity through IL-10-producing B cells

Summary: Prior exposure to pathogen-associated stimuli can reduce morbidity and mortality to new infections through innate immune training, which can be further improved by childhood vaccination. This study demonstrates that inflammatory training of fibroblastic reticular cells (FRC) in the gut enhances immune responses to bacterial pathogens and reduces colon inflammation through IL-10 production by B cells, ultimately improving clearance of infection. These findings suggest a new mode of immune training through FRC that can impact future adaptive responses and better preserve host health.

Dendrobium fimbriatum polysaccharide ameliorates DSS-induced intestinal mucosal injury by IL-22-regulated intestinal stem cell regeneration

Summary: In this study, the intervention effect of Dendrobium fimbriatum Hook polysaccharide (cDFPW1) on ulcerative colitis (UC) was confirmed through experiments on mice. The results showed that cDFPW1 significantly alleviated UC symptoms and promoted the proliferation of intestinal epithelial cells. Furthermore, cDFPW1 directly improved intestinal organoid damage and increased the number of epithelial cells, promoting stem cell-mediated intestinal epithelial regeneration.

PD-1 and TIM-3 differentially regulate subsets of mouse IL-17A-producing γδ T cells

Summary: This study reveals that different subsets of IL-17A-producing gamma delta T cells in the lung of mice are controlled by PD-1 and TIM-3 in steady-state and cancer. Vγ6(+) cells express high levels of PD-1, while Vγ4(+) cells upregulate TIM-3 in response to tumor-related stimulation. Inhibition of PD-1 or TIM-3 increases the numbers of Vγ6(+) and Vγ4(+) cells respectively. Moreover, genetic deletion of gamma delta T cells enhances responsiveness to anti-PD-1 and anti-TIM-3 immunotherapy in a mammary tumor model, indicating that IL-17A-producing gamma delta T cells contribute to immunotherapy resistance.

Anti-ICOS mAb Targets Pathogenic IL-17A-expressing Cells in Canine Model of Chronic GVHD

Summary: Chronic graft-versus-host disease (GVHD) is a significant issue in transplant patients, and targeting specific T-cell populations like CD4(+) and CD8(+) T cells may be crucial in developing effective treatments for the disease. Treatment with anti-inducible costimulator (ICOS) monoclonal antibodies showed promising results in transiently relieving symptoms and prolonging survival in dogs with chronic GVHD, particularly targeting IL-17A(+) cells, but further research is needed to control both CD4(+) and CD8(+) T cells for better outcomes.

Results showed that the novel CAR NK cell is functional and further investigations are warranted.

Summary: Despite the effectiveness of immune checkpoint inhibitors (ICIs) in lung cancer, there is a lack of knowledge about predictive biomarkers. This study aimed to analyze different subsets of T-lymphocytes and natural killer (NK) cells as predictive biomarkers in a cohort of patients with nonsmall cell lung cancer (NSCLC) treated with ICI. The results showed that a subset of circulating CD56+CD16-PD-1+ NK cells have a good predictive ability, providing valuable information for precision medicine protocols.

Summary: Sirolimus, an inhibitor of mTOR, shows potential as a component of GVHD prophylaxis regimens in HSCT, but detailed immunologic studies are lacking. This study evaluated the effect of sirolimus on immune reconstitution using longitudinal patient samples. Sirolimus inhibited naive cell suppression and NK cell reconstitution, leading to impaired proliferation and functional responses.
