Congenital heart malformation (CHM) is the most common form of congenital human birth anomaly and is the leading cause of infant mortality. Although some causative genes have been identified, little progress has been made in identifying genes in which low-penetrance susceptibility variants occur in the majority of sporadic CHM cases. To identify common genetic variants associated with sporadic non-syndromic CHM in Han Chinese populations, we performed a multistage genome-wide association study (GWAS) in a total of 4,225 CHM cases and 5,112 non-CHM controls. The GWAS stage included 945 cases and 1,246 controls and was followed by 2-stage validation with 2,160 cases and 3,866 controls. The combined analyses identified significant associations (P < 5.0 x 10(-8)) at 1p12 (rs2474937 near TBX15; odds ratio (OR) = 1.40; P = 8.44 x 10(-10)) and 4q31.1 (rs1531070 in MAML3; OR = 1.40; P = 4.99 x 10(-12)). These results extend current knowledge of genetic contributions to CHM in Han Chinese populations.

Structure-Property Relationship of Cyano-Functionalized Conjugated Polymers for Photocatalytic Hydrogen Production

Xu Chi, Qian Chen, Zhi-An Lan, Xirui Zhang, Xiong Chen, Xinchen Wang

Summary: Conjugated polymers (CPs) have received increasing attention in photocatalysis due to their stability and molecular tunability. In this study, a new efficient cyano (CN) engineering approach was proposed to enhance the photocatalytic performance of CPs. Three CP-based photocatalysts with different CN contents were synthesized to investigate the relationship between CN functionalization and photocatalytic activity. Experimental and theoretical studies revealed that CN functionalization strengthened the donor-acceptor (D-A) interaction, enhanced light absorption, charge separation/transfer efficiency, hydrophilicity of CPs, and facilitated the output of photoinduced electrons from CPs to Pt cocatalyst.

Summary: The expression of flavonoid synthesis-related genes in Panax notoginseng stems shows multi-peak curves, and the distribution of total flavonoid and anthocyanin content in the stems is asymmetric, mainly concentrated in the upper parts. The correlations between gene expression and flavonoid accumulation are highly diverse.

Liu Yang, Lufang Feng, Lijuan Huang, Xuejiao Li, Wenwen Qiu, Kehu Yang, Jie Qiu, Haihong Li

Summary: This study conducted a meta-analysis to identify the association between maternal factors and intrauterine growth retardation (IUGR), and found that smoking, primiparity, prepregnancy BMI < 18.5, poor gestational weight gain, pregnancy-induced hypertension, anemia, and hypoproteinemia were associated with IUGR. These findings could contribute to the development of risk prediction models and improve management and education for child-bearing or early pregnant women.

A novel protein encoded by circRsrc1 regulates mitochondrial ribosome assembly and translation during spermatogenesis

Shu Zhang, Chang Wang, Yue Wang, Hao Zhang, Chen Xu, Yiwei Cheng, Yan Yuan, Jiahao Sha, Xuejiang Guo, Yiqiang Cui

Summary: In this study, an endogenous circular RNA (circRsrc1) was discovered, which encodes a novel protein (Rsrc1-161aa). Deletion of Rsrc1-161aa in mice resulted in impaired male fertility, characterized by decreased sperm count and motility due to mitochondrial energy metabolism dysfunctions. In vitro experiments showed that circRsrc1 regulates mitochondrial functions through its encoded protein Rsrc1-161aa. Mechanistically, Rsrc1-161aa interacts with mitochondrial protein C1qbp to enhance its binding activity to mitochondrial mRNAs, thereby regulating mitochondrial ribosome assembly and translation.

Multi-Omics Approaches for Revealing the Epigenetic Regulation of Histone H3.1 during Spermatogonial Stem Cell Differentiation In Vitro

Li Liu, Haojie Li, Mengjie Wang, Xiangzheng Zhang, Jie Ren, Yan Yuan, Jiahao Sha, Xuejiang Guo

Summary: Epigenetic regulation, specifically histone post-translational modifications (PTMs), is involved in the differentiation of spermatogonial stem cells (SSCs). However, due to the low number of SSCs in vivo, there is a lack of systematic studies on the regulation of histone PTMs during SSCs differentiation. In this study, we used targeted quantitative proteomics with mass spectrometry and RNA-seq data to quantify the dynamic changes of 46 different PTMs on histone H3.1 during SSCs differentiation in vitro. We identified seven differentially regulated histone H3.1 modifications and further identified specific binding proteins for H3K9me2 and H3S10ph, including transcription factors GTF2E2 and SUPT5H, which appear to play a crucial role in the epigenetic regulation of SSC differentiation.

DeepSP: A Deep Learning Framework for Spatial Proteomics

Bing Wang, Xiangzheng Zhang, Chen Xu, Xudong Han, Yue Wang, Chenghao Situ, Yan Li, Xuejiang Guo

Summary: The study introduces a new deep learning framework, DeepSP, for protein subcellular localization (PSL) prediction using mass spectrometry-based spatial proteomics data. By capturing detailed changes in protein occupancy profiles across subcellular fractions, DeepSP constructs a new feature map and utilizes a convolutional block attention module to enhance PSL prediction performance. DeepSP demonstrates significant improvement in accuracy and robustness compared to current machine learning predictors, making it a valuable tool for spatial proteomics studies and the understanding of protein function.

Summary: In this article, six novel heterocyclic Ir(III) complexes were designed and calculated using DFT and TD-DFT. The bands of the lowest-lying absorptions and lowest energy emissions, as well as the spectral properties, frontier molecular orbitals, and spin-orbital coupling values, were studied. These complexes exhibit low efficiency roll-off properties, which is significant for the development of OLEDs.
