Emerimicins V-X, 15-Residue Peptaibols Discovered from an Acremonium sp. through Integrated Genomic and Chemical Approaches

Summary: The fermentation of Acremonium tubakii W. Gams isolated from soil samples collected from the University of Utah led to the discovery of six new linear pentadecapeptides, named emerimicins V-X. Detailed bioinformatic investigations and experimental techniques were used to elucidate the amino acid composition and configuration of these peptides, revealing significant activity in two of them.

Summary: Natural products like conotoxins have great potential in biomedical research and treating human diseases. Developing a rapid screening method using zebrafish larvae, researchers identified a bioactive conotoxin, CNF-Ep1, which not only affected the locomotor behavior of the larvae but also showed activity in mouse assay systems. This novel screening platform can accelerate the discovery of bioactive marine natural products with potential implications for understanding neuronal circuitry.

MIC-Drop: A platform for large-scale in vivo CRISPR screens

Summary: MIC-Drop is a platform combining droplet microfluidics, single-needle en masse CRISPR ribonucleoprotein injections, and DNA barcoding, enabling large-scale functional genetic screens in zebrafish to efficiently identify genes responsible for morphological or behavioral phenotypes. It has the potential to scale to thousands of genes, facilitating genome-scale reverse genetic screens in model organisms.

Rapid Mounting of Zebrafish Larvae for Brain Imaging

Summary: This study developed a method using a computer numerical controlled manufactured mold to create larva-shaped slots in agarose gel for easier mounting of zebrafish larvae in a vertical position. Social behavioral profiling by unsupervised deep learning reveals a stimulative effect of dopamine D3 agonists on zebrafish sociality

Summary: Researchers developed the ZeChat assay, a scalable social behavioral evaluation method for zebrafish using unsupervised deep learning. Screening a compound library, they found that different classes of chemicals induce distinct patterns of social behavior. Dopamine D3 agonists were found to have a social stimulative effect and rescued social deficits in a zebrafish autism model. The ZeChat platform offers a promising approach for studying the pharmacology of social behavior and discovering new social-modulatory compounds.
