Stilbene production in cell cultures of Vitis vinifera L. cvs Red Globe and Michele Palieri elicited by methyl jasmonate

Cell cultures obtained from Vitis vinifera cvs Michele Palieri and Red Globe were cultured in order to stimulate stilbene production. In the calli, stilbene production peaked at day 22 of culture for both cultivars; the main compound was trans-piceid, followed by cis-piceid. Methyl jasmonate, which was added to cell suspensions in the first half of the exponential growth phase, enhanced stilbene accumulation, producing mainly trans-piceid and epsilon-viniferin. Other stilbenoids, though in lower quantities, were identified by liquid chromatography/positive electrospray mass spectrometry. epsilon-Viniferin and trans-resveratrol were the main compounds released into the culture medium. The total quantity of stilbenes was genotype dependent, with a better response found for the cv Red Globe.

Summary: Research on wild neotropical blueberries, endemic to Central and South American regions, has been limited despite their potential as sources of bioactive compounds. This study focused on the phenolic composition of Disterigma alaternoides for the first time, identifying 249 phenolic compounds including anthocyanins, flavonoids, phenolic acids, and proanthocyanidins. The findings highlight the importance of further exploring and understanding the diverse phenolic compounds present in these uncharted matrices.

Investigating the Short Peptidome Profile of Italian Dry-Cured Ham at Different Processing Times by High-Resolution Mass Spectrometry and Chemometrics

Summary: In this study, a suspect screening approach based on high-resolution mass spectrometry was used to comprehensively characterize the short endogenous peptidome in dry-cured ham samples. A total of 236 short peptide sequences, including 173 natural short peptides and 63 sequences containing non-proteinogenic amino acids, were tentatively identified. Several modified short peptides, such as pyroglutamination and lactoyl conjugation, were also observed, suggesting reactions occurring over time.

Stereoselective Synthesis of Spiro-Decalin Oxindole Derivatives via Sequential Organocatalytic Michael-Domino Michael/Aldol Reaction

Summary: A highly stereoselective procedure for the synthesis of spiro-polycyclic oxindoles bearing five contiguous stereogenic centers including two tetrasubstituted carbons has been developed. The sequence involves sequential organocatalysis using a pyrrolidine-based organocatalyst and DBU, resulting in an atom-economical Michael-domino Michael/aldol reaction sequence. The reaction yielded variously functionalized spiro-decalin oxindoles with excellent stereoselectivity (>99:1 dr, up to 92% ee).

Summary: This paper proposes a method of using 6-azauracil for the nitrogenous Paterno-Buchi reaction of lipids to determine carbon-carbon double bonds in fatty acyl chains using higher collisional dissociation in the negative ion mode. The method improves the ionization efficiency of fatty acid precursors and diagnostic product ions, and also allows for deprotonation of phosphatidylcholines.

Summary: In this study, a multiplexed strategy combining a nanoparticle-enabled blood test and serum levels of acute-phase proteins demonstrated the ability to distinguish pancreatic cancer patients from healthy controls. The study suggests the potential role of acute-phase proteins as biomarkers for pancreatic cancer and provides a new avenue for the development of multiplexed technologies for early cancer detection.

Xanthones: Biosynthesis and Trafficking in Plants, Fungi and Lichens

Summary: Xanthones are valuable secondary metabolites produced by plants with diverse structures and biological activities. This review summarizes the current knowledge of xanthone biosynthesis in plants, including the precursors, enzymes involved, and their cellular and organ localization. The biosynthetic pathway in plants involves the shikimate and acetate pathways, leading to the central intermediate 2,3',4,6-tetrahydroxybenzophenone. Unlike plants, xanthones in fungi and lichens are entirely derived from polyketide. Further studies are needed to investigate the synthesis mechanisms in order to exploit xanthones for practical applications.

New hydrophilic material based on hydrogel polymer for the selective enrichment of intact glycopeptides from serum protein digests

Summary: The paper presents the preparation and characterization of a new HILIC material for the enrichment of N-linked glycopeptides. The material was prepared using specific monomer and cross-linker, and by fine-tuning the polymer properties, it displayed superior performance in terms of the number of enriched glycopeptides and selectivity. These results not only provide a new HILIC material for glycopeptide enrichment but also pave the way for the use of hydrogel materials in this field.

Evaluation of the carotenoid and fat-soluble vitamin profile of industrial hemp inflorescence by liquid chromatography coupled to mass spectrometry and photodiode-array detection

Summary: Industrial hemp is being widely used for pharmaceutical and nutraceutical purposes, but the detailed composition of hemp remains poorly understood. In this study, the carotenoids and fat-soluble vitamins in the inflorescences of seven Italian industrial hemp varieties were analyzed. The most abundant carotenoids were found to be phytoene, phytofluene, and all-trans -a-carotene. In addition, several other carotenoids were tentatively identified using liquid chromatography coupled with photodiode-array detection.

Biotic transformation products of sulfonamides in environmental water samples: High-resolution mass spectrometry-based tentative identification by a suspect screening approach

Summary: The presence of pharmaceuticals in the aquatic environment is mainly due to their release from wastewater treatment plants. Sulfonamides, a class of synthetic antibacterials, have often been found in surface water, contributing to antibiotic resistance. Monitoring their transformation products (TPs) is important, as they may still exert pharmaceutical activity. In this study, three commonly used sulfonamides were incubated with activated sludge, and their TPs were identified using mass spectrometry. The identified TPs were then used to analyze wastewater treatment plant samples. Acetylation was found to be the predominant transformation pathway.

A Lupinus angustifolius protein hydrolysate exerts hypocholesterolemic effects in Western diet-fed ApoE-/- mice through the modulation of LDLR and PCSK9 pathways

Summary: This study elucidated the molecular mechanism underlying the hypocholesterolemic effects of Lupin protein hydrolysates (LPHs) in an in vivo model. The results showed that LPH treatment reduced de novo cholesterol synthesis through the regulation of HMGCoAR and LDLR protein levels. LPHs also controlled the PSCK9 signal pathway, leading to decreased cholesterol absorption. These findings highlight LPHs as a potential ingredient for developing new functional foods.
