Summary: We designed a hybrid system with a polymeric core surrounded by a shell of different lipids to combine the excellent transfection properties of lipids with the high stability of polymeric nanoparticles. By adjusting the amount of the zwitterionic phospholipid DOPE in the lipid layer, we improved the cell tolerability of the nanocarriers. LPN(70/30) showed the best transfection performance.

Summary: This study aimed to develop surface modified PLGA nanocarriers to protect a protein-based antigen in the stomach and release it at target intestinal sites. The surface modified PLGA nanoparticles showed better protection of the antigen in simulated gastric and intestinal fluids compared to non-functionalized nanoparticles. The surface modified nanoparticles exhibited sustained release of the antigen and maintained its structural integrity. Overall, this study demonstrates the safety and potential of surface modified PLGA nanoparticles for oral protein-based vaccine delivery.

Summary: Allergen immunotherapy (AIT) is the only curative treatment for allergic diseases, but it has limitations. In this study, we developed a new approach using MAN-coated ovalbumin (OVA) nanoparticles (MDO) to target dendritic cells (DCs) more effectively and promote higher antigen presentation efficiency. The results showed that DCs exposed to MDO induced Treg cells and MDO had low reactivity and high safety. In a mouse model of allergic asthma, MDO had significant preventative and therapeutic effects.

Summary: An immunization experiment using PLGA nanoparticles encapsulating inactivated NDV vaccine was conducted in specific pathogen-free chickens. The results showed that the nano-NDV vaccine induced high levels of antibodies and cell-mediated immune responses, providing 100% protection against NDV challenge. This study provides insights for developing PLGA nanoparticle-based vaccines against avian diseases.

Summary: This study investigates the role of CRIF1 deficiency in modulating mitochondrial oxidative phosphorylation (OXPHOS) system dysfunction and its antitumor effects in MCF-7 breast cancer cells. CRIF1 silencing decreases the assembly of mitochondrial OXPHOS complexes, leading to mitochondrial dysfunction and elevated reactive oxygen species (ROS) levels. Inhibition of CRIF1 suppresses cell proliferation and inhibits cell migration in MCF-7 cells, and intratumoral injection of CRIF1 siRNA nanoparticles inhibits tumor growth in MCF-7 xenograft mice.

Summary: Nanoparticle-based mRNA cancer vaccines show great promise for personalized cancer treatments. We developed a bioreducible lipophilic poly(beta-amino ester) nanocarrier that can efficiently deliver multiple antigen-encoding mRNAs and nucleic acid-based adjuvants to dendritic cells. The engineered nanocarrier facilitated targeted delivery without the need for surface functionalization and resulted in robust CD8+ T cell responses and efficient antitumor therapy in murine melanoma and colon adenocarcinoma models.

Summary: In this study, PLGA-based curcumin nanoparticles modified with folic acid and chitosan (curcumin-PLGA-CS-FA) were prepared and their effects on the redox status of glioblastoma tumor cells were evaluated. The results showed that curcumin nanoparticles could modulate the antioxidant-oxidant balance and might be effective in the prevention and treatment of glioblastoma.

The unique product ions generated from the different functional groups in these compounds allow for specific identification and development of quantification methods using MS/MS technology.
