Summary: There is a pressing need for the development of highly sensitive and selective fluorescence probes for ONOO- in mitochondria. In this study, we introduced a ratiometric fluorescent probe COUS, which utilized a coumarin-cyanine hybrid as the fluorophore and C = C bonds as reaction sites of ONOO-. The probe COUS displayed sensitivity and selectivity towards ONOO-, with a large fluorescence emission shift (239 nm) and a low detection limit (41.88 nM). Furthermore, COUS demonstrated mitochondrial targeting ability, and the targeting moiety could dissociate from the probe upon reaction with ONOO-, enabling accurate detection of ONOO- in mitochondria.

Summary: In this study, a novel negative photochromic spiropyran derivative, MAHD-SP, was designed with stable ring-opened state and notable orange fluorescence. The fluorescence of MAHD-SP was quenched by the Michael addition reaction of SO2 derivative. A ratiometric fluorescent polymeric nanoprobe (RFPN) was constructed using MAHD-SP, a fluorescent conjugated polymer PFO, and a polymeric surfactant PEO113-b-PS49. The RFPN exhibited high sensitivity and selectivity for the ratiometric detection of SO2 derivatives in aqueous solutions, and could visualize SO2 derivative generation in living cells.

Summary: Near infrared (NIR) light has high transparency in biological tissue, and recent advances in NIR fluorophores have enabled effective use of the NIR optical window for in vivo bioimaging and photodynamic therapy. This review focuses on the development of hybrid nanostructures by utilizing the polarity and vibrational energy of ions and molecules surrounding the luminescence centers, with the aim of reducing heat emission and achieving bright luminescence. The guidelines proposed in the review are based on the multiphonon relaxation theory and emphasize the importance of surrounding luminescence centers with ions with low vibrational energy or molecules with small chemical polarity.

Summary: This study developed a one-step hydrothermal method to synthesize biocompatible fluorescent carbon quantum dots (CQDs) from Aegle marmelos fruit extract. The CQDs had a naturally distributed size, unintentional doping with heteroatoms, and exhibited strong photostability and water solubility. The study also explored the variables affecting CQD photoluminescence and demonstrated the potential of Ca, K, N-doped CQDs as fluorescent biomarkers for bio-imaging applications, with satisfactory cell survival and cytotoxicity results.

Summary: The liver, as the metabolic and detoxification center, plays a vital role in various physiological processes. Normal liver function is crucial for maintaining overall health, while abnormal liver function can lead to liver diseases. Early diagnosis of liver diseases is essential for timely treatment. The use of fluorescent probes for detecting biomarkers of liver diseases has provided valuable insights for pathological studies, drug development, and disease diagnosis and treatment in recent years. This review summarizes the progress of fluorescence probes in detecting hepatopathy-related biomarkers and discusses the current challenges and future directions. The development of long wavelength probes, multi-detection probes, quantitative analysis probes, and multi-modal probes is expected to be a popular trend. This review aims to facilitate the development of novel fluorescent probes for disease detection and provide guidance for diagnosis and treatment.
