Summary: By conducting a case-control study, we found that Parkinson's disease (PD) patients do not commonly have an increase in resting energy expenditure (REE), but it does increase in patients with obesity and more severe disease. The accuracy of existing REE equations is limited, while the new equation we proposed in this study provides better REE estimates.

Summary: This study used a two-sample Mendelian randomization method to analyze the associations of smoking, alcohol drinking, and coffee drinking with Parkinson's disease (PD). The results showed that smoking was inversely associated with PD, while genetic liability was positively associated with PD. The associations of alcohol drinking and coffee drinking with PD were limited by insufficient statistical power.

Gut Microbiota in Monozygotic Twins Discordant for Parkinson's Disease

Summary: Differences in gut microbiota between Parkinson's disease patients and controls may be influenced by various confounding factors. Using monozygotic twins, this study controlled for these factors and found minimal differences in bacterial taxa abundance and predicted metabolic pathways. Further research is needed to understand the role of gut microbiota in the pathogenesis of Parkinson's disease.

Summary: This study conducted a large-scale independent replication of the interaction between HLA-DRB1 and smoking in Parkinson's disease. The valine at position 11 (V11) in HLA-DRB1 showed the strongest association with PD at the amino acid level. The results suggest an inverse association between genetically predicted smoking initiation and PD, particularly in individuals without the V11 variant. In silico predictions support this interaction pattern by showing the influence of V11 and smoking-induced modifications on binding affinity of alpha-synuclein.

Precision Medicine in Parkinson's Disease: From Genetic Risk Signals to Personalized Therapy

Summary: Understanding the pathophysiology and genetic background of Parkinson's disease is crucial for developing effective disease-modifying therapeutic strategies. The discovery of genetic variants causing or increasing the risk for PD has contributed to refining the classification of the disease and guiding the decision-making process on different therapies. However, the heterogeneity in molecular pathophysiology among PD populations may explain the disappointing results of current disease-modifying strategies. Personalized medicine should be prioritized in the future. This review discusses the current therapeutic options for PD and the challenges in successful clinical trials, as well as the role of genetics in guiding treatment decisions in daily clinical practice.
