BackgroundLow serum uric acid (UA) levels have been associated with increased Parkinson's disease (PD) risk and accelerated disease progression. We analyzed the effect of polymorphisms in 9 genes influencing serum UA concentration on the risk of PD. MethodsWe genotyped SLC2A9 rs734553, ABCG2 rs2231142, SLC17A1 rs1183201, SLC22A11 rs17300741, SLC22A12 rs505802, GCKR rs780094, PDZK1 rs12129861, LRRC16A+SCGN rs742132, and SLC16A9 rs12356193 in 1061 PD patients and 754 controls. For each subject we calculated a cumulative genetic risk score (GRS), defined as the total number of PD-risk alleles (range, 2-15) associated to lower serum UA levels. Serum UA levels were measured in a subgroup of 365 PD cases and 132 controls. ResultsSerum UA levels were significantly lower in men with PD than in controls. Subjects (both men and women) carrying more than 9 risk alleles (third GRS tertile) had a 1.5 higher risk of developing PD than subjects with less than 8 risk alleles (first GRS tertile). An inverse correlation was observed between higher GRS and lower serum UA concentration in both men and women. Summary: This study analyzed the frequency, causes, and predictors of acute hospitalization in a cohort of Parkinson's disease patients in Spain. The results showed that falls were the most common cause of hospitalization and an independent predictor of acute hospitalization in Parkinson's disease patients.

Investigating the effects of COVID-19 lockdown on Italian children and adolescents with and without neurodevelopmental disorders: a cross-sectional study

Summary: A cross-sectional study compared the effects of social distancing and lifestyle changes during the COVID-19 lockdown on children and adolescents with and without Neurodevelopmental Disorders (NDDs). Results showed that NDDs significantly impacted remote learning, sociality, scolding, and anxiety levels. There was agreement between parents and children regarding remote learning, lifestyle, and scolding. This study highlights the importance of supportive strategies for youth with NDDs during periods of lockdown.

Summary: Mosaic loss of chromosome Y (mLOY) is a common somatic event associated with Alzheimer's disease (AD) and can be age-confounded in genome-wide association studies. This study used Mendelian randomisation to construct an age-independent mLOY polygenic risk score (mloy-PRS) and found that higher genetic risk for mLOY was associated with faster progression to AD in men with mild cognitive impairment. The study suggests that mLOY is involved in AD pathogenesis and shows associations with cerebrospinal fluid biomarkers.

Summary: Visual hallucinations and subjective cognitive complaints in Parkinson's disease are associated with cognitive impairment. This study aimed to determine the association between visual hallucinations and cognitive complaints and the risk of developing cognitive impairment in Parkinson's disease patients with normal cognition. The results showed that visual hallucinations were associated with the development of cognitive impairment, and the presence of both visual hallucinations and cognitive complaints increased the probability of developing cognitive impairment.

Summary: This study applied the MNCD classification to Parkinson's disease patients and analyzed its correlation with quality of life and disease severity. The results showed that a more advanced MNCD stage was associated with poorer quality of life and greater disease severity. Therefore, MNCD could be a useful tool for monitoring disease progression in Parkinson's disease.
