Aqueous Fraction of Nephelium ramboutan-ake Rind Induces Mitochondrial-Mediated Apoptosis in HT-29 Human Colorectal Adenocarcinoma Cells

The aim of this study was to investigate the cytotoxic and apoptotic effects of Nephelium ramboutan-ake (pulasan) rind in selected human cancer cell lines. The crude ethanol extract and fractions (ethyl acetate and aqueous) of N. ramboutan-ake inhibited the growth of HT-29, HCT-116, MDA-MB-231, Ca Ski cells according to MTT assays. The N. ramboutan-ake aqueous fraction (NRAF) was found to exert the greatest cytotoxic effect against HT-29 in a dose-dependent manner. Evidence of apoptotic cell death was revealed by features such as chromatin condensation, nuclear fragmentation and apoptotic body formation. The result from a TUNEL assay strongly suggested that NRAF brings about DNA fragmentation in HT-29 cells. Phosphatidylserine (PS) externalization on the outer leaflet of plasma membranes was detected with annexin V-FITC/PI binding, confirming the early stage of apoptosis. The mitochondrial permeability transition is an important step in the induction of cellular apoptosis, and the results clearly suggested that NRAF led to collapse of mitochondrial transmembrane potential in HT-29 cells. This attenuation of mitochondrial membrane potential (Delta psi m) was accompanied by increased production of ROS and depletion of GSH, an increase of Bax protein expression, and induced-activation of caspase-3/7 and caspase-9. These combined results suggest that NRAF induces mitochondrial-mediated apoptosis.

Nano-engineered ZnO/CNF-based epoxidized natural rubber with enhanced strength for novel Self-healing glove fabrication

Summary: This study successfully fabricates rubber gloves with excellent mechanical properties and self-healing ability by synthesizing zinc oxide-cellulose nanofiber (ZnO-CNF) reinforced epoxidized natural rubber (ENR) composite. The addition of ZnO-CNF filler significantly improves the tensile strength and thermal stability of the gloves. The resulting ENR/ZnO-CNF gloves exhibit high strength, strain recovery, and thermo-responsive self-healing behavior.

The Convergence of FTIR and EVs: Emergence Strategy for Non-Invasive Cancer Markers Discovery

Summary: In conjunction with imaging analysis, pathology-based assessments of biopsied tissue are the gold standard for diagnosing solid tumors. However, the disadvantages of tissue biopsies have led to the development of liquid biopsy, which involves sampling and clinical assessment of bodily fluids for cancer diagnosis. Extracellular vesicles (EVs), carrying molecular profiles of their cell or tissue origins, have emerged as one of the most promising biomarkers for cancer, allowing for non-invasive and rapid diagnosis with the help of Fourier-transform infrared spectroscopy (FTIR).

Mentha longifolia Essential Oil and Pulegone in Edible Coatings of Alginate and Chitosan: Effects on Pathogenic Bacteria in Lactic Cheese

Summary: This study examined the antibacterial effects of Mentha longifolia essential oil and pulegone in edible coatings on the growth of Staphylococcus aureus, Listeria monocytogenes, and Escherichia coli in cheese. The results showed that the addition of these compounds significantly reduced bacterial growth during storage. Increasing the concentration of chitosan, M. longifolia, and pulegone in the coatings further decreased the bacterial population, with pulegone exhibiting a stronger effect. Overall, the findings suggest that M. longifolia essential oil and pulegone, combined with alginate and chitosan coatings, have antibacterial effects against S. aureus, L. monocytogenes, and E. coli in cheese.

Diabetes Mellitus and Its Therapy in Brunei Darussalam-A Critical Revisit

Summary: This study aimed to identify and review research related to the practice of patients with DM in Brunei Darussalam in order to identify associated factors influencing diabetic medication knowledge and adherence. The results revealed relatively poor medication knowledge and inappropriate diabetes-related practices in studies conducted in Brunei Darussalam. However, no study has directly measured both medication awareness and compliance among Brunei Darussalam T2DM patients. Future studies can focus on the relation between these two factors in different geographical locations characterized by different diabetes-related practices and culture.

Summary: This study investigated the suitability of 3D cultures as cell-based models for GBM by leveraging genomic data. Genes related to pathways such as epithelial-to-mesenchymal transition, angiogenesis/migration, hypoxia, stemness, and the Wnt signalling pathway were found to be upregulated in both brain samples from GBM patients and 3D GBM cells. These findings support the use of 3D GBM cultures as reliable preclinical models for studying GBM.
