Summary: A novel cholinesterase gene from the larvae of Amblyomma americanum was identified and confirmed to be expressed in both tick synganglion and saliva. The recombinant enzyme showed high sensitivity to eserine and BW284c51, and hydrolyzed BuTCh faster than ATCh. The presence of the antigenic protein in tick saliva suggests potential roles in hydrolyzing cholinergic substrates and modulating host immune responses to tick feeding and pathogens.

Summary: Previous studies have shown that the EF-1 & alpha; promoter in episomal vectors maintains high-level transgene expression in CHO cells, but further improvement is needed. In this study, we incorporated MARs, UCOE, and STAR 40 elements into the episomal vector at different sites and orientations and evaluated their effects on transgene expression in CHO-K1 cells. The results demonstrated that MAR X-29 upstream of the promoter and MAR1 downstream of the poly A significantly increased eGFP expression, and the combination of MAR X-29 and hCMVI resulted in the highest transgene expression levels. Additionally, the vector with MAR X-29+hCMVI induced HSV-TK protein expression and showed cell-killing and bystander effects on HCT116 cells.
