Summary: The combination of the sphingosine analog FTY-720 and the mitochondrial Kv1.3 inhibitor PAPTP can induce death of pancreatic cancer cells by inhibiting acid sphingomyelinase and disrupting mitochondrial and lysosomal functions. This study reveals a previously unknown crosstalk between lysosomes and mitochondria mediated by sphingolipid metabolism.

Summary: This review summarizes the effects of overexpression of antioxidant system genes on stress resistance and carcinogenesis. In most cases, overexpression of these genes has been shown to increase cellular and organism resistance to oxidative and genotoxic stress, but has different effects on cancer initiation and promotion. Therefore, the prospects of using this approach in practice are greatly limited.

A particular lipidome signature was identified in patients with different outcomes after 7 days, with an increase in specific lipid species like ceramides and sulfatides, potentially serving as biomarkers for disease progression and severity.

Summary: Overexpression of S-R protein in Schizochytrium leads to improved cell growth and significant increase in total fatty acid content, as well as upregulated production of 0-carotene. The overexpression of S-R protein also enhances hypersaline tolerance in Schizochytrium, resulting in increased dry cell weight and production of glycerol and proline. This study provides a feasible strategy for engineering Thraustochytriaceae for efficient biomass and biochemical production.

Summary: Aberrant activation of Ras-ERK signaling pathway drives important cancer phenotypes. Inhibition of this pathway by natural compound betulinic acid (BA) showed anti-cancer effects in NSCLC cells, with enhanced efficacy when combined with ERK inhibitors. However, the combination also increased protective autophagy and AKT phosphorylation, suggesting a complex interaction between these pathways in cancer therapy. The triple-drug combination of BA, ERKi, and autophagy inhibitor HCQ demonstrated superior therapeutic efficacy in vivo in a mouse model of NSCLC.

Summary: The overexpression of VaPYL4 gene enhances the cold, drought, and salt tolerance in grape and Arabidopsis plants. Transgenic plants show higher survival rates, earlier flowering, and heavier biomass. Physiological analysis reveals lower content of malondialdehyde and higher peroxidase activity in the transgenic plants. Therefore, VaPYL4 gene plays an important role in plant growth and development under adverse environments.
