Novel Locally Active Estrogens Accelerate Cutaneous Wound Healing. A Preliminary Study

New 17 beta-estradiol (E-2) derivatives 1-11 were synthesized from an estrone derivative by addition of organometallic reagents prepared from protected alpha,omega-alkynols and further elaboration of the addition products. The estrogenic activity of these novel compounds was determined using in vitro binding competition assay and transactivation analysis. Among the E-2 derivatives synthesized, compound 2 showed the highest transactivation potency and was therefore tested for its ability to modulate cutaneous wound healing in vivo. Compound 2's ability to accelerate wound healing in ovariectomized mice and decrease the production of inflammatory molecules was comparable to that of E-2. However, the activity of compound 2 was not superimposable to E-2 with regard to the cells involved in the wound repairing process. When locally administered, compound 2 did not show any systemic activity on ER. This class of compounds with clear beneficial effects on wound healing and suitable for topical administration may lead to the generation of innovative drugs for an area of unmet clinical need.

Hypothalamic NPY-Y1R Interacts with Gonadal Hormones in Protecting Female Mice against Obesity and Neuroinflammation

Summary: Conditional inactivation of the Npy1r gene in forebrain principal neurons leads to a sexually dimorphic phenotype in mice, with male mice showing more significant effects than females. Additionally, the conditional deletion of the Npy1r gene in male mice increases susceptibility to obesity when exposed to a high-fat diet. In female mice, the conditional deletion of the Npy1r gene increases body weight and subcutaneous white adipose tissue weight in both standard diet and high-fat diet-fed ovariectomized females, possibly mediated by the NPY-Y1R's ability to protect the brain against neuroinflammation.

Summary: Naked mole-rats show similar skin aging characteristics to humans, including a decrease in epidermal thickness and keratinocyte proliferation. However, interestingly, the expression levels of several longevity-associated and tumor-suppressor genes are increased in aged naked mole-rat skin. These findings provide a basis for using naked mole-rats as a model organism to study the biology and disease resistance of human skin.

Stereoselective Synthesis of Spiro-Decalin Oxindole Derivatives via Sequential Organocatalytic Michael-Domino Michael/Aldol Reaction

Summary: A highly stereoselective procedure for the synthesis of spiro-polycyclic oxindoles bearing five contiguous stereogenic centers including two tetrasubstituted carbons has been developed. The sequence involves sequential organocatalysis using a pyrrolidine-based organocatalyst and DBU, resulting in an atom-economical Michael-domino Michael/aldol reaction sequence. The reaction yielded variously functionalized spiro-decalin oxindoles with excellent stereoselectivity (>99:1 dr, up to 92% ee).

Oleochemical carbonates: A comprehensive characterization of an emerging class of organic compounds

Summary: Dialkyl carbonates with short-medium alkyl length, also known as oleochemical carbonates, have attractive properties and a wide range of applications. However, their chemical physical properties and microscopic correlations are not well understood. In this study, we explored the temperature dependence of these properties and the microscopic morphology of DACs. Our findings reveal a distinct degree of mesoscopic spatial segregation and the role of carbonate moieties in driving weak hydrogen bonding interactions and alkyl chain alignment. These findings have implications for the potential applications of DACs in separation, catalysis, and lubrication fields.

N-Acetyl-l-phenylalanine Racemization during TBTU Amidation: An In-Depth Study for the Synthesis of Anti-Inflammatory 2-(N-Acetyl)-l-phenylalanylamido-2-deoxy-d-glucose (NAPA)

Summary: A study was conducted on the amidation conditions of N-acetyl-l-phenylalanine using TBTU and various bases for the synthesis of NAPA, a potential drug for joints diseases. The diastereoselective amidation reaction with 1,3,4,6-tetra-O-acetyl-beta-d-glucosamine showed racemization, but the use of pyridine as base showed potential for reducing racemization. Stereochemical retention under amidation conditions was studied in detail through control experiments.

Summary: In the last decade, there has been an increasing number of studies on the application of dynamic covalent chemistry in polymeric materials, due to the unique properties of these materials such as reprocessability, recyclability, and self-healing. This research focuses on the synthesis and characterization of a dynamic covalent chemistry-based polymer, which is reprocessable and self-healable. The polymer also shows strong interaction with copper surfaces through stable Cu-S bonds. Furthermore, the study investigates its potential application as a solvent-free reusable adhesive for copper.

Sex-Specific Microglial Responses to Glucocerebrosidase Inhibition: Relevance to GBA1-Linked Parkinson's Disease

Summary: Microglia are heterogenous cells in the nervous system that have distinct populations contributing to specific processes, including neuroprotection. Our study found that male microglia tend to have a more pro-inflammatory phenotype, while female microglia are more sensitive to glucocerebrosidase inhibition. Furthermore, glucocerebrosidase inhibition impaired the ability of female microglia to enhance the Nrf2-dependent detoxification pathway in neurons.
