Summary: Extrinsic apoptosis is mediated by the activation of death receptors and involves the assembly of the death-inducing signaling complex (DISC). This process includes multiple steps such as dimerization of procaspase-8 and its processing, which can be carefully monitored using the described experimental workflow.

Summary: By designing a peptide probe and reduced graphene oxide, a new biosensing system was constructed for accurate detection and intracellular imaging of caspase-8 activity. The proposed method exhibited a linear response range and low detection limit in vitro, and successfully monitored the upregulation of intracellular caspase-8 activity.

Summary: This study found that connexin32 (Cx32) is associated with necroptosis in HCC cells, mediating the activation of necroptosis by binding to Src. Experimental results suggest that necroptosis inducers may be a favorable strategy for eliminating HCC cells with high expression of Cx32.

Summary: Regulated cell death is essential in development and cellular homeostasis. The core FADD:Caspase-8 complex and its regulatory partners, such as the cell death inhibitor c-FLIP, play a critical role in coordinating cell fate. The recruitment of c-FLIPS into this complex inhibits Caspase-8 activity by altering its structure.

Summary: In this study, oat bran was enriched with blackcurrant and blueberry powder to form paste extracts, which were found to significantly inhibit the growth and invasion of hepatocellular carcinoma cells. Mechanistic studies suggested that these extracts induced cell apoptosis through cell cycle arrest and regulation of apoptotic-related protein expressions. The extracts also increased reactive oxygen species levels in the cancer cells via the Nrf2/HO1 signaling pathway. These findings highlight the therapeutic potential of blueberry and blackcurrant-enriched oat bran paste for patients with liver cancer.

Summary: The interaction between TRAIL and IER3 can induce apoptotic death of HCC cells and inhibit their proliferation and migration by inhibiting Wnt/beta-catenin signaling. This TRAIL/IER3/beta-catenin axis may serve as a viable therapeutic target for HCC patients.

Summary: The Drosophila tumour necrosis factor (TNF) system consists of a single ligand, Eiger (Egr), and two receptors. The crystallographic structure of Egr in complex with the extracellular domain of the receptor Grindelwald suggests that distinct affinities of TNF ligand for its receptors mediate non-redundant functions.

Summary: In this study, lentivirus-mediated Gag-CASP8 successfully delivered activated CASP8 into HER-2-overexpressing primary human breast cancer cells to induce apoptosis by activating caspase-3. This therapeutic approach can block cell proliferation and migration, providing a new avenue for tumor gene therapy.

Summary: Liver cancer is the sixth most prevalent and the third most fatal type of cancer worldwide. Conventional anticancer drugs have limited efficacy due to their short half-life, poor solubility, and inefficient drug delivery. Therefore, it is crucial to develop novel treatment strategies, such as using exosomes as delivery agents, to overcome these obstacles and achieve effective and long-lasting anticancer treatments.
