Metabolomics study on model rats of chronic obstructive pulmonary disease treated with Bu-Fei Jian-Pi

The therapeutic effect of Traditional Chinese Medicine (TCM) on chronic obstructive pulmonary disease (COPD) has been know for numerous years; however, the mechanism of action of the beneficial effects of TCM remains to be elucidated. The present study aimed to investigate the molecular mechanisms of COPD through metabolomic analysis as well as explore the targets and intervention mechanisms of TCM therapy using the common TCM granules Bu-Fei Jian-Pi. COPD rat models were established using smoke inhalations and recurrent bacterial infections. Rats were then divided into three groups as follows: A1, control healthy rats; B1, COPD model; and D1, Bu-Fei Jian-Pi-treated COPD rats. Following administration of the medicine, the metabolomic profile of the lung tissue of rats in each group was assessed using high-performance liquid chromatography/quadrupole-time-of-flight mass spectrometry. The results demonstrated that there was a significanlty different spectrum of metabolites in the lung tissue of the model group compared to that of the control group as well as the Bu-Fei Jian-Pi-treated COPD group; in addition, following treatment with Bu-Fei Jian-Pi, the metabolites of COPD rats were comparable with those of the control. Notable changes were observed in 31 metabolites between the Bu-Fei Jian-Pi-treated group and the model group; however, there were 13 comparable metabolites between the Bu-Fei Jian-Pi and control groups as well as the model and control groups. Eleven metabolites showed a negative fold change in the Bu-Fei Jian-Pi-treated groups compared to concentrations in the model group; however, minimal changes were observed in phenylpyruvic acid and alpha-D-fucose expression. In conclusion, the results of the present study demonstrated that Bu-Fei Jian-Pi granules had beneficial effects on measured outcomes in a rat model of stable COPD, indicated by a significantly different spectrum of metabolites. This therefore indicated that the metabolites which had significantly altered expression in the model group compared with that of the control and Bu-Fei Jian-Pi-treated groups may be potential biomarkers of COPD.

A Novel and Reliable Rat Model of Autism

Summary: This study established an autism rat model by transplanting fecal sample extract of ASD children into pregnant rats, showing typical behavioral characteristics of ASD. The gut microbiota of the model mainly consisted of Firmicutes and Bacteroidetes, reflecting the abnormal gut microbiota of ASD patients. The model displayed high structural validity and potential as a new and reliable animal model for studying ASD.

Discovery of grey matter lesion-related immune genes for diagnostic prediction in multiple sclerosis

Summary: This study investigated the gray matter lesion-related genes in multiple sclerosis (MS) and identified TLR9, CCL5, CXCL8, and PDGFRB as potential biomarkers for gray matter injuries in MS. A reliable prediction model was established, and gene enrichment analysis revealed their association with cell adhesion molecules, cytokine-cytokine receptor interaction, and the JAK-STAT signaling pathway. These findings provide guidance for therapeutic intervention in MS.

Downregulation of FTO in the hippocampus is associated with mental disorders induced by fear stress during pregnancy

Summary: Mental disorders during pregnancy, such as anxiety and depression, can lead to adverse outcomes, but the underlying mechanisms are still being investigated. This study found that the FTO protein, which is involved in RNA modification, plays a role in and regulates mental disorders induced by fear stress in pregnant rats. The study also identified specific genes that are directly regulated by FTO and may be involved in the development of mental disorders.

Integrated bioinformatic analysis and experimental validation for exploring the key molecular of brain inflammaging

Summary: This study integrated bioinformatics and experimental validation to explore the mechanisms of inflammaging in the brain. The research found that inflammatory-related genes were closely associated with brain aging, and animal studies were conducted to validate the selected biomarkers. The study also discovered that CX3CL1 may play a role in the development of inflammaging by regulating macrophages, which suggests that targeting CX3CL1 can potentially delay inflammaging and immunosenescence in the brain.
