Assessment of immune responses of the flagellin (FliC) fused to FimH adhesin of Uropathogenic Escherichia coli

Urinary tract infection (UTI) caused by Uropathogenic Escherichia coli (UPEC) is one of the most common infectious diseases in the world. Despite extensive efforts, a vaccine that protects humans against UTI is currently missing. In this study, the immunogenicity of flagellin (FliC) of UPEC strain in different vaccine combinations with FimH antigen of OPEC and conventional adjuvant Montanide ISA 206 was assessed. Finally, efficacy of the immune responses was evaluated for protection of the bladder and kidney of challenged immunized mice. Mice immunized with the fusion FimH.FliC induced significantly higher anti-FliC humoral (IgG1) and cellular (Th1 and Th2) immune responses than with FliC alone or FliC admixed with FimH. The Montanide enhanced the immune responses of FliC antigen and directed the anti-FliC responses preferentially toward Th1. The FliC vaccine combinations reduced bladder infection as compared to control mice. The fusion FimH.FliC and FliC admixed with FimH and Montanide combinations gave the best results in protection of kidney infection, compared to the control mice. The results of this study propose new promising vaccine combinations based on the FliC antigen and Montanide against UTI caused by UPEC. (C) 2012 Elsevier Ltd. All rights reserved.

Computational evaluation of modified peptides from human neutrophil peptide 1 (HNP-1)

Summary: The development of bacterial resistance to antibiotics has led to the attention of antimicrobial peptides (AMPs). This study computationally investigated the interaction between derived peptides and bacterial membrane and found that the derived peptides could exhibit similar antimicrobial effects to HNP-1. Furthermore, chemical modifications improved the physicochemical properties of the peptides and retained similar binding affinity.

Summary: This study investigated new strategies to enhance the immune response to HIV-1 vaccine using immunological adjuvants. The results showed that conjugating HIV-1 p24-Nef with type-A flagellin significantly increased lymphocyte proliferation and cytokine production, leading to improved humoral and cellular immune responses. Furthermore, the route of vaccine administration also affected the immune response.

Summary: This study evaluated the antimicrobial effects of peptide IDR1018 and chitosan nanoparticles (CNs) against ciprofloxacin-resistant uropathogenic Escherichia coli (UPEC). The results showed that peptide IDR1018 and CNs have antimicrobial properties on UPEC isolates and can inhibit and eradicate biofilms in a concentration-dependent manner.

Detection of ESBL and AmpC producing Klebsiella pneumoniae ST11 and ST147 from urinary tract infections in Iran

Summary: In this study, 200 Klebsiella pneumoniae isolates were collected from patients with urinary tract infections in Tehran, Iran. The isolates were analyzed for antibiotic resistance, ESBLs, AmpCs, integron genes, plasmids, and genomic relatedness. The results showed high levels of resistance against ampicillin, with most isolates carrying beta-lactamase genes and the most prevalent integron being class I. The study also identified the replicons of plasmids and the ability of selected isolates to transfer genes. Finally, the isolates were categorized into pulsotypes and MLST identified sequence types.

Different Formulations of Inactivated SARS-CoV-2 Vaccine Candidates in Human Compatible Adjuvants: Potency Studies in Mice Showed Different Platforms of Immune Responses

Summary: In this study, different formulations of the inactivated SARS-CoV-2 virus were developed with different adjuvants, and the immune responses were assessed. The results showed that Montanide 51VG and Montanide ISA720VG adjuvants were more effective in inducing immune responses compared to Alum adjuvant.

Circulating LncRNAs landscape as potential biomarkers in breast cancer

Summary: This study used qRT-PCR to measure the expression levels of lncRNAs in breast cancer patients and healthy women. The results showed that certain circulating lncRNAs were down-regulated in breast cancer patients, while others were up-regulated. SPRY4-IT1, XIST, and H19 lncRNAs showed excellent potential in distinguishing breast cancer from healthy controls. Furthermore, H19 and SPRY4-IT1 lncRNAs were found to be significantly correlated with clinicopathological traits in breast cancer patients.

The Effect of Ophthalmic and Systemic Formulations of Latilactobacillus sakei on Clinical and Immunological Outcomes of Patients With Dry Eye Disease: A Factorial, Randomized, Placebo-controlled, and Triple-masking Clinical Trial

Summary: This study evaluated the efficacy and safety of Latilactobacillus sakei as an ophthalmic bacterial lysate or oral probiotic on patients with dry eye disease (DED). Results showed that the ophthalmic drops significantly decreased DED signs and symptoms, while the oral capsules had no effect. The ophthalmic formulation containing L. sakei lysate improved DED and suppressed ocular surface inflammatory response.

Formulation of Recombinant H1N1 Hemagglutinin in MF59 and Alum Adjuvants: A Comparison of the Vaccines Potency and Efficacy in BALB/C Mice

Summary: In this study, the expression and potency of a recombinant H1N1 hemagglutinin (HA) vaccine were evaluated in a mouse model. The vaccine, produced in an SF9 cell line and formulated in MF59 adjuvant, showed a robust immune response and cytotoxic T lymphocyte (CTL) activity compared to an alum-based vaccine and phosphate-buffered saline (PBS). Additionally, the MF59-based vaccine demonstrated superior humoral immune responses and increased survival rates in comparison to the alum-based vaccine. The MF59 formulation seems to contribute to the superiority of the candidate vaccine.

This study aimed to design a cross-reactive epitope vaccine to stimulate CD8 + and CD4 + T cells against UTI pathogens. Predictions based on informatics were used to design potentially cross-protective T-cells inducing vaccine candidates.
