HLJ1 deficiency leads to reduced IL-12 levels, dampened IFN-gamma production in natural killer cells, and improved survival rate. HLJ1 converts misfolded IL-12p35 homodimers to monomers, maintaining bioactive IL-12p70 heterodimerization and secretion.

This study reveals a novel role for A20 in regulating IL-33-induced STAT1 signaling in macrophages.

Summary: This study demonstrates the important role of IL-23/IL-23R in modulating intracellular bacterial clearance in macrophages during mycobacterial infection. IL-23/IL-23R enhances bacterial clearance, T helper cell differentiation, and proinflammatory cytokine secretion, thereby increasing host bactericidal activity. IL-23/IL-23R could serve as potential targets for the prevention and treatment of leprosy and other mycobacterial infections.

Summary: This study presents a case series of four pediatric patients with Mendelian susceptibility to mycobacterial diseases (MSMD), specifically with IFN gamma R1 and STAT1 deficiencies, and cutaneous non-tuberculous mycobacteria (NTM)/bacillus Calmette-Guerin (BCG) infections. The results highlight the importance of considering MSMD in patients with seemingly normal immune function who present with NTM/BCG infections, as timely diagnosis and treatment can significantly improve disease outcomes.

Monoclonal antibodies targeting IL-12 and/or IL-23 have been developed for the treatment of Crohn's disease and ulcerative colitis. This Review summarizes the biological characteristics of IL-12 and IL-23, discusses their role in intestinal homeostasis and inflammation, and highlights the development of drugs targeting IL-12 and/or IL-23.

Summary: This study aimed to characterize the mutations in genes of the IL-12/23/ISG15-IFN-gamma pathway in severe tuberculosis patients. Through the investigation of four clinically suspected tuberculosis patients, mutations were identified in IL-12R beta 1, STAT1, NEMO, and CYBB genes, which are associated with the onset of tuberculosis and immune dysfunction.

Summary: This study investigates the mechanisms by which the interaction between infectious agent molecules and early innate responses enhances the early inhibition of cancer growth, and specifically evaluates the role of Interleukin-12 (IL-12). The results show that CpG-protected mice against the early growth of plasmacytoma TEPC.1033.C2 cells through the ligation of toll-like receptor 9 (TLR9). This protection is dependent on IL-12 and gamma-interferon, and is confirmed in a model of early mesothelioma AB1 cell growth through the stimulation of TLR3 and TLR7/8.

Summary: Inflammatory bowel disease, including Crohn's disease and ulcerative colitis, is a group of complex immune-mediated disorders. Targeting the IL-23/Th-17 axis has shown effectiveness in the treatment of these diseases. IL-12/23 inhibitors have been approved for both Crohn's disease and ulcerative colitis, while IL-23p19 antagonists have shown better outcomes in psoriasis patients. Many specific IL-23p19 antagonists are being studied in the treatment of inflammatory bowel disease.

The fusion of IL-12 with diabodies successfully achieved binding to tumor cells and reduced its systemic availability. Summary: This study evaluated the association of SNPs in innate-immunity and MMP genes with clinical outcomes in patients with chronic Q fever. The results showed that SNP rs3751143 in P2RX7 was associated with therapy failure, while SNP rs7125062 in MMP1 was associated with fewer chronic Q fever-related complications such as acute aneurysms.
