Bile Acid Receptor Agonist GW4064 Regulates PPARγ Coactivator-1α Expression Through Estrogen Receptor-Related Receptor α

Peroxisome proliferator-activated receptor alpha coactivator-1 alpha (PGC-1 alpha) is induced in energy-starved conditions and is a key regulator of energy homeostasis. This makes PGC-1 alpha an attractive therapeutic target for metabolic syndrome and diabetes. In our effort to identify new regulators of PGC-1 alpha expression, we found that GW4064, a widely used synthetic agonist for the nuclear bile acid receptor [farnesoid X receptor (FXR)] strongly enhances PGC-1 alpha promoter reporter activity, mRNA, and protein expression. This induction in PGC-1 alpha concomitantly enhances mitochondrial mass and expression of several PGC-1 alpha target genes involved in mitochondrial function. Using FXR-rich or FXR-nonexpressing cell lines and tissues, we found that this effect of GW4064 is not mediated directly by FXR but occurs via activation of estrogen receptor-related receptor alpha (ERR alpha). Cell-based, biochemical and biophysical assays indicate GW4064 as an agonist of ERR proteins. Interestingly, FXR disruption alters GW4064 induction of PGC-1 alpha mRNA in a tissue-dependent manner. Using FXR-null [FXR knockout (FXRKO)] mice, we determined that GW4064 induction of PGC-1 alpha expression is not affected in oxidative soleus muscles of FXRKO mice but is compromised in the FXRKO liver. Mechanistic studies to explain these differences revealed that FXR physically interacts with ERR and protects them from repression by the atypical corepressor, small heterodimer partner in liver. Together, this interplay between ERR alpha-FXR-PGC-1 alpha and small heterodimer partner offers new insights into the biological functions of ERR alpha and FXR, thus providing a knowledge base for therapeutics in energy balance-related pathophysiology. Dexamethasone activates c-Jun NH2-terminal kinase (JNK) which interacts with GR and protects it from ubiquitin-mediated degradation in NSCLC cells

Summary: Dexamethasone treatment leads to an increase in glucocorticoid receptor (GR) protein levels in non-small cell lung cancer (NSCLC) cells through JNK activation. JNK phosphorylation precedes GR upregulation, and JNK-mediated GR stabilization affects its nuclear transcriptional functional activity in NSCLC cells. Increased levels of both JNK and GR contribute towards better prognosis of NSCLC patients.

Ormeloxifene, a nonsteroidal antifertility drug promotes megakaryocyte differentiation in leukemia cell line K562

Summary: ORM can induce megakaryocytic and myeloid differentiation in the K562 cell line. This induction is mediated by activation of ERK and induction of autophagy. Additionally, inhibition of caspases can mitigate megakaryocytic differentiation. These findings suggest that ORM has potential as an inducer of megakaryocytic differentiation.

AIP4 regulates adipocyte differentiation by targeting C/EBPa for ubiquitin-mediated proteasomal degradation

Summary: In this study, it was found that E3 ubiquitin ligase AIP4 negatively regulates C/EBPa protein stability, leading to reduced adipogenesis. AIP4 overexpression inhibits lipid accumulation in 3T3-L1 preadipocytes, while AIP4 depletion promotes lipid accumulation even in the absence of differentiation-inducing media. AIP4 physically interacts with C/EBPa and promotes its ubiquitination, resulting in proteasomal degradation.

Bisphenol-A (BPA) Impairs Hippocampal Neurogenesis via Inhibiting Regulation of the Ubiquitin Proteasomal System

Summary: Injectable hydrogels have been used for cartilage repair, but some of them have issues with rapid degradation and low mechanical strength. To address these challenges, a bioinspired double network hydrogel (BDNH) was developed, which exhibits temperature-dependent stiffening after implantation. The BDNH showed excellent cell viability, long time cell proliferation, and cartilage specific matrix production, making it a potential candidate for cartilage tissue engineering.
