Esophageal squamous cell carcinoma (ESCC) is one of the most common tumors in the world. It is very urgent to develop new therapeutic strategies. MED23, a component of the mediator complex, is known as a hub to integrate various signaling pathways. However, the function of MED23 in ESCC remains unknown. Here, we found that the expression of MED23 was downregulated in the clinical ESCC samples and the expression of MED23 reversely correlated with tumor size and clinical stage. Moreover, overexpression of MED23 in ESCC cells inhibited cell growth dramatically, while downregulation of MED23 promoted the tumorigenecity of ESCC cells in vitro and in vivo. Mechanistically, knockdown the expression of MED23 inhibited cell apoptosis by downregulation of Bax, activated Caspase 3, activated Caspase 9 and upregulation of cyclinD1 and Bcl2. Taken together, our study revealed the suppressive role of MED23 in ESCC and MED23 might be an important therapeutic target in ESCC. (c) 2013 Wiley Periodicals, Inc.

Four hub genes regulate tumor infiltration by immune cells, antitumor immunity in the tumor microenvironment, and survival outcomes in lung squamous cell carcinoma patients

Tuo Zhang, Haitang Ang, Beibei Sun, Feng Yao

Summary: This study identified hub genes regulating tumor infiltration by immune cells and antitumor immunity in lung squamous cell carcinoma (LUSC) through bioinformatics analyses. It also discovered different immune subgroups in LUSC tissues based on ssGSEA and ESTIMATE analyses, showing significant differences in tumor purity.

Dysregulated Glutamate Transporter SLC1A1 Propels Cystine Uptake via Xc- for Glutathione Synthesis in Lung Cancer

Summary: SLC1A1, a glutamate transporter, plays a crucial role in cystine uptake and GSH biosynthesis in lung cancer cells, impacting tumor development. Dysregulated SLC1A1 enhances cystine uptake, while depletion inhibits it, leading to oxidative stress-induced cell death or growth inhibition. Glutamate transporters provide additional therapeutic targets for cancer treatment beyond Xc(-) regulation.

NF2 and Canonical Hippo-YAP Pathway Define Distinct Tumor Subsets Characterized by Different Immune Deficiency and Treatment Implications in Human Pleural Mesothelioma

Haitang Yang, Sean R. R. Hall, Beibei Sun, Liang Zhao, Yanyun Gao, Ralph A. Schmid, Swee T. Tan, Ren-Wang Peng, Feng Yao

Summary: The study revealed that loss-of-function in the upstream regulator NF2 of the Hippo pathway is associated with aberrant activation of Hippo-YAP-independent signaling, defining distinct MPM subsets with different molecular features and prognosis. This has important clinical implications for precision oncology in MPM patients.
