Summary: This study presents an ultrasound-mediated nanobubble (NB)-based gene delivery system for treating bone disorders. The system utilizes NBs as controlled-release carriers for delivering a mixture of CTSK siRNA and CeNPs, aiming to downregulate CTSK expression in osteoclasts and enhance bone formation. The release profiles of CTSK siRNA and CeNPs were measured after low-intensity pulsed ultrasound (LIPUS) stimulation, and the effects were evaluated using staining techniques. The study provides a potential method for gene delivery and treatment of bone disorders.

Summary: Precise detection of early osteolytic metastases remains challenging due to limited sensitivity and specificity of traditional imaging techniques. A fluoro-photoacoustic dual-modality imaging probe has been developed, consisting of a near-infrared dye connected to a cathepsin K (CTSK)-cleavable peptide sequence and functionalized with osteophilic alendronate. In vitro and in vivo experiments demonstrate that the probe generates both near-infrared fluorescent and photoacoustic signals in response to CTSK, offering a potential strategy for detecting deep-seated early osteolytic metastases.
