Summary: This study reveals that high levels of HDGF are associated with poor survival prognosis in breast cancer, and its upregulation contributes to radioresistance in breast cancer cells. Depletion of HDGF can enhance the sensitivity of radioresistant breast cancer cells to drugs targeting STAT3 phosphorylation. These findings suggest that HDGF may serve as a potential molecular target for predicting radioresistance in breast cancer.

Summary: Cancer cells develop radioresistance in lung adenocarcinoma radiotherapy, and this study reveals that upregulated expression of FIBP protein is associated with worse overall survival in lung adenocarcinoma. Depletion of FIBP inhibits cancer progression and radioresistance by interacting with STAT3 and enhancing its transcriptional activity to induce the expression of EME1. The findings suggest that targeting FIBP may be a novel intervention strategy for lung adenocarcinoma radiotherapy.

Summary: This study found that overexpression of KDM4C is associated with poor prognosis in lung cancer patients; genetic or pharmacological inhibition of KDM4C can reduce tumorigenesis and radioresistance in lung cancer; USP9X-mediated deubiquitination of KDM4C activates TGF-beta 2/Smad signaling pathway to promote radioresistance in lung cancer.

Summary: Currently, recurrence and metastasis remain significant factors leading to poor prognosis in colorectal cancer (CRC) patients. Cancer-associated fibroblasts (CAFs) play a crucial role in promoting tumorigenesis and development. Bufalin, the main active monomer of the clinical drug cinobufacini, has exhibited anti-tumor activity in various cancers. However, limited research has investigated the effect of bufalin in inhibiting metastasis from the perspective of the tumor microenvironment. This study isolated CAFs from freshly resected CRC patient specimens and observed their effect on CRC cell invasion. The study also explored the physiological activity of CRC mediated by CAFs and the inhibitory effect of bufalin on CAF-mediated CRC invasion and metastasis through the STAT3 pathway.

Summary: This study identified a novel mechanism in which CTSH signaling regulates radioresistance in HCC cells. Disrupting CTSH signaling was found to impair the high glycolysis metabolism of tumor cells, enhance aerobic respiration, and promote apoptosis through upregulation and release of proapoptotic factors such as AIFM1, HTRA2, and DIABLO, consequently reducing radioresistance.

Summary: The engineered therapeutic protein PAK has been found to reverse multidrug resistance in cancer cells, enhancing their sensitivity to chemotherapy. PAK induces mitochondrial injury by selectively inhibiting mitochondrial complex V, leading to redistribution of ABC transporters and reversal of drug resistance.

Summary: Ovarian cancer is a highly lethal malignancy, often diagnosed at advanced stages due to the lack of early diagnosis methods. The standard treatment includes surgery and chemotherapy, but many patients develop chemoresistant tumors. Therefore, there is a need to develop new therapeutic agents to overcome chemoresistance. Niclosamide, an anti-parasite agent, has shown potent anti-cancer activities in ovarian cancer and other types of human cancers. This study investigates the potential of repurposing niclosamide as a therapeutic agent.

Summary: SERPINE2 is identified as a regulator of radiosensitivity and the DNA damage response (DDR) in lung cancer, with knockdown improving tumor radiosensitivity by reducing homologous recombination repair, restoring cell cycle checkpoints, and suppressing migration and invasion. Additionally, high SERPINE2 expression correlates with poor prognosis and a high serum concentration predicts a poor response to radiotherapy in lung cancer patients.

Summary: This study found that STAT3 can directly activate the pentose-phosphate pathway, exerting a pro-oncogenic effect on Taxol resistance in ovarian cancer. Inhibiting STAT3 can decrease G6PD expression and enhance Taxol sensitivity in resistant cells. This research provides a potential therapeutic target for improving the prognosis of ovarian cancer patients.

Summary: The study identified PKP2 as a critical driver of radiation resistance in lung cancer, with high PKP2 expression predicting poor survival rates post-radiotherapy. Mechanistically, PKP2 was found to be methylated at the arginine site and interacted with PRMT1, leading to stabilization of beta-catenin and induction of LIG4-mediated repair. Treatment with the PRMT1 inhibitor C-7280948 abolished PKP2-induced radioresistance, suggesting its potential as a radiosensitizer in lung cancer.

Summary: This study found that the upregulation of PPDPF in NSCLC correlated with poor overall survival. PPDPF promoted the growth, colony formation, and invasion of lung cancer cells. Mechanistically, it interacted with BABAM2, blocking its ubiquitination and stabilizing BABAM2, leading to radioresistance in lung cancer cells.

Summary: In this study, a novel benzothiazole derivative PB01 was shown to have antitumor effects in radiation-resistant human NSCLC cells by inducing cell death and inhibiting epithelial-to-mesenchymal transition. The mechanism involved induction of reactive oxygen species, ER stress, and activation of the ATR axis, suggesting that PB01 treatment could overcome radio-resistance during NSCLC radiotherapy.
