Summary: Most ovarian cancer patients develop recurrent cancers which are often resistant to commonly employed chemotherapy agents, such as cisplatin. In this study, we investigated the role of heat shock protein 27 (HSP27) and fatty acid oxidation (FAO) in promoting cisplatin resistance. Our results showed that inhibiting HSP27 and FAO enhanced the cytotoxic effect of cisplatin and depleted cisplatin-induced ROS in cisplatin-resistant ovarian cancer cell lines. This dual inhibition strategy also increased cisplatin's cytotoxicity in in vivo xenograft tumors.

Summary: Tumor-specific alterations in metabolism are crucial for sustaining cell growth and survival in various cancers, but the heterogeneous nature of metabolism poses challenges for effective anticancer therapies. Medulloblastoma is a highly heterogeneous pediatric brain tumor with complex intratumoral cellular diversity. This review highlights the mechanisms through which medulloblastoma cells rewire their metabolism and energy production networks to support rapid growth, resistance to stress, invasion, metastasis, and therapy. The potential clinical benefits of targeting energy metabolism in suppressing medulloblastoma progression and enhancing current therapies are also discussed.

Summary: This study evaluated the super relaxed state of myosin and sarcomeric protein phosphorylation in two pathological models of cardiomyopathy and a near-physiological model of cardiac hypertrophy. The mutations in the cardiomyopathy models had antagonistic effects on the myosin energetic states, leading to changes in RLC phosphorylation levels. The N-terminus of ELC was suggested to act as an energetic switch promoting the transition between SRX and DRX states, affecting myosin RLC phosphorylation.
