Targeting Olfactomedin-like 3 Inhibits Tumor Growth by Impairing Angiogenesis and Pericyte Coverage

Antiangiogenic drugs have been used as anticancer agents to target tumor endothelial cells or pericytes. Because of limited efficacy of the current monotherapies, there is a strong demand for the dual targeting of endothelial cells and pericytes. Here, we identify Olfactomedin-like 3 (Olfml3) as a novel proangiogenic cue within the tumor microenvironment. Tumor-derived Olfml3 is produced by both tumor endothelial cells and accompanying pericytes and deposited in the perivascular compartment. Blockade of Olfml3 by anti-Olfml3 antibodies is highly effective in reducing tumor vascularization, pericyte coverage, and tumor growth. In vitro, Olfml3 targeting is sufficient to inhibit endothelioma cell migration and sprouting. Olfml3 alone or through binding to BMP4 enhances the canonical SMAD1/5/8 signaling pathway required for BMP4-induced angiogenesis. Therefore, Olfml3 blockade provides a novel strategy to control tumor growth by targeting two distinct cell types within the tumor microenvironment using a single molecule. Summary: Endothelial cells play a crucial role in cardiovascular health, and dysfunction can be caused by cardiovascular risk factors but restored once stimuli are removed. There are various methods to measure endothelial function, with each having its pros and cons. It is recommended to adopt a consensus methodology like flow-mediated dilatation and establish reference values for different populations. Future studies should explore the integration of endothelial function measurements with patient-specific epigenetic data and other biomarkers.

Summary: The study reveals that the catecholamine noradrenaline plays a role in the response of infantile hemangioma (IH) to propranolol treatment. IH tissues contain a higher amount of noradrenaline compared to involuted IHs or propranolol-treated IHs. The sensitivity of IH to propranolol depends on the crosstalk between endothelial cells, pericytes, and telocytes in the presence of a high local amount of noradrenaline.

Translational opportunities of single-cell biology in atherosclerosis

Summary: The advent of single-cell biology has opened up new possibilities for understanding human biological processes and diagnosing, monitoring, and treating diseases, including cardiovascular disease (CVD). By analyzing CVD samples at the single-cell level, new cell communities that play a role in disease development can be identified, leading to new therapeutic strategies. This review focuses on the single-cell analysis of atherosclerotic plaques, highlighting the current understanding of cellular subpopulations and their heterogeneity and plasticity in relation to clinically relevant features. The clinical impact of single-cell technologies in CVD patient care is emphasized, calling for multidisciplinary and international collaborative efforts.

Atorvastatin-loaded spray-dried PLGA microparticles for local prevention of intimal hyperplasia: Drug release rate optimization and activity on synthetic vascular smooth muscle cells

Summary: This study aimed to encapsulate a high payload of atorvastatin (ATV) to prevent intimal hyperplasia (IH) in vascular bypass grafts and evaluate its impact on synthetic smooth muscle cells (SMCs) involved in IH progression. Poly(lactic-co-glycolic acid) microparticles (MPs) were formulated with different drug loadings and tested for ATV release, sterilization, and effects on SMCs. The results showed sustained ATV release from 10% and 20% ATV-loaded MPs for 90 days. The ATV-loaded MPs reversed the synthetic phenotype of SMCs and attenuated their migratory and proliferative capacities.

Neutralization of S100A4 induces stabilization of atherosclerotic plaques: role of smooth muscle cells
