Molecular Evolutionary Analysis of Cancer Cell Lines

With genome-wide cancer studies producing large DNA sequence data sets, novel computational approaches toward better understanding the role of mutations in tumor survival and proliferation are greatly needed. Tumors are widely viewed to be influenced by Darwinian processes, yet molecular evolutionary analysis, invaluable in other DNA sequence studies, has seen little application in cancer biology. Here, we describe the phylogenetic analysis of 353 cancer cell lines based on multiple sequence alignments of 3,252 nucleotides and 1,170 amino acids built from the concatenation of variant codons and residues across 494 and 523 genes, respectively. Reconstructed phylogenetic trees cluster cell lines by shared DNA variant patterns rather than cancer tissue type, suggesting that tumors originating from diverse histologies have similar oncogenic pathways. A well-supported clade of 91 cancer cell lines representing multiple tumor types also had significantly different gene expression profiles from the remaining cell lines according to statistical analyses of mRNA microarray data. This suggests that phylogenetic clustering of tumor cell lines based on DNA variants might reflect functional similarities in cellular pathways. Positive selection analysis revealed specific DNA variants that might be potential driver mutations. The results show that a significant number of parents and school-aged children reported the elimination of outdoor play, especially in low-income families. Additionally, the elimination of outdoor activity is associated with child obesity.

Engineered microbial systems for advanced drug delivery

Complex genetic modifications in live bacteria and bacteria-derived particles simulate lipid-based vectors and enable the delivery of therapeutic molecules. The potential translation of these drug delivery systems into clinical therapies is discussed.

Summary: This study provides comprehensive insights into the changes in lipid levels during pregnancy and early childhood, as well as their association with obesity risk. The findings suggest that the lipidomic profiles in pregnant and non-pregnant women, as well as in offspring at birth and at 6 years of age, are influenced by adiposity. The study also highlights the overlap in the circulating lipid phenotype of obesity risk between children and adults, with a larger number of lipids associated with BMI in adults.

The results show that the diet quality of preschool-aged children is generally low, with some excessive intake. Summary: This study examined the effects of the timing of introduction of complementary foods and the duration of breastfeeding on body fat partitioning and cardiometabolic risk in children. The results showed that shorter breastfeeding duration and earlier introduction of complementary foods were associated with increased child adiposity and cardiometabolic risk. Longer duration of breastfeeding may protect against the cardiometabolic risk associated with early introduction of complementary foods.

Nutrient trajectories during infancy and their associations with childhood neurodevelopment

Summary: The purpose of this study was to examine the associations between infants' dietary nutrient trajectories and subsequent neurodevelopment during childhood. One-day food records were collected at ages 6, 9, and 12 months, and neurodevelopment assessments were conducted at ages 24 and 54 months. The findings showed that higher protein intake and increasing rate of intake were associated with higher fine motor scores at 24 months, while higher fat intake was associated with higher receptive language scores. However, no significant associations were observed with neurodevelopment at 54 months. Overall, this study provides important insights into the relationship between nutrition and child neurodevelopment over time.

Quality of life and psychosocial outcomes among children with metabolically healthy and unhealthy obesity

Summary: This study compared health-related quality of life (HRQoL) and psychosocial outcomes between children with metabolically healthy obesity (MHO) and metabolically unhealthy obesity (MUO) using validated questionnaires. The results showed that children with MUO and prior knowledge of existing metabolic conditions had lower HRQoL, higher depression, and stress than children with MHO. Additionally, the metabolic health status of fathers was found to influence HRQoL in children with MUO.

Summary: Adopting a healthy diet during and after pregnancy is important for women's cardiometabolic health. Changes in diet quality from pregnancy to 6 years postpregnancy were related to cardiometabolic markers 8 years postpregnancy in a study of 652 women. Improving or preventing a decline in diet quality postpregnancy may improve lipid profile and insulin resistance.

Summary: This study examined longitudinal changes in physical activity and sedentary behavior patterns from preconception to postpartum. The findings showed that walking time increased during late pregnancy, while vigorous-intensity physical activity and moderate to vigorous physical activity decreased but partially returned to preconception levels postpartum. Sedentary time remained stable during pregnancy but decreased postpartum. Individual characteristics such as ethnicity, body mass index, employment, parity, and self-rated general health significantly influenced women's activity patterns.
