Shotgun proteomics reveals specific modulated protein patterns in tears of patients with primary open angle glaucoma naive to therapy

Primary open angle glaucoma (POAG) is one of the main causes of irreversible blindness worldwide. The pathogenesis of POAG is still unclear. Alteration and sclerosis of trabecular meshwork with changes in aqueous humor molecular composition seem to play the key role. Increased intraocular pressure is widely known to be the main risk factor for the onset and progression of the disease. Unfortunately, the early diagnosis of POAG still remains the main challenge. In order to provide insight into the pathophysiology of glaucoma, here we report a shotgun proteomics approach to tears of patients with POAG naive to therapy. Our proteomics results showed 27 differential tear proteins in POAG vs. CTRL comparison (25 up regulated proteins in the POAG group and two unique proteins in the CTRL group), 16 of which were associated with inflammatory response, free radical scavenging, cell-to-cell signaling and interaction. Overall the protein modulation shown in POAG tears proves the involvement of biochemical networks linked to inflammation. Among all regulated proteins, a sub-group of 12 up-regulated proteins in naive POAG patients were found to be down-regulated in medically controlled POAG patients treated with prostanoid analogues (PGA), as reported in our previous work (i.e., lipocalin-1, lysozyme C, lactotransferrin, proline-rich-protein 4, prolactin-inducible protein, zinc-alpha-2-glycoprotein, polymeric immunoglobulin receptor, cystatin S, Ig kappa chain C region, Ig alpha-2 chain C region, immunoglobulin J chain, Ig alpha-1 chain C region). In summary, our findings indicate that the POAG tears protein expression is a mixture of increased inflammatory proteins that could be potential biomarkers of the disease, and their regulation may be involved in the mechanism by which PGA are able to decrease the intraocular pressure in glaucoma patients.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a leading cause of cancer mortality worldwide. This study investigated the crosstalk between PDAC cells and associated fibroblasts. Proteomic analysis was used to identify proteins released from PDAC cells and detect proteostatic changes in normal fibroblasts treated with PDAC conditioned medium. The results suggest that the activation of migration, vasculogenesis, cellular homeostasis, metabolism of amino acids, and reduced apoptosis may be regulated by ITGB3 and TGFB1/2 followed by SMAD3, STAT3, and BAG3 activation.

Summary: Recent studies have focused on the protective and/or pathological role of virus-specific T cells in SARS-CoV-2 infection. This study found that the immune response produced by natural infection is similar to that induced by vaccination. Summary: Prevention of cardiovascular disease with low-dose aspirin may be less effective in patients with faster recovery of platelet cyclooxygenase-1 activity. Patients with accelerated COX-1 recovery exhibit reduced platelet glycoprotein Ib alpha shedding, leading to increased thrombopoietin production and the formation of platelets that are not inhibited by aspirin over 24 hours. Clinical markers including age, non-alcoholic fatty liver disease, visceral obesity, and TPO/GC ratio can accurately predict faster COX-1 recovery and suboptimal aspirin response. The TPO/GC ratio may serve as a simple tool to identify patients who would benefit from more frequent aspirin dosing.

Skin tests combined with the basophil activation test (BAT) using PEG2000, PEG4000, and DMG-PEG2000 can help evaluate allergic reactions to the vaccine. The study suggests that even when the BAT is negative, a high stimulation index (SI) in basophil activation may indicate a low-grade reaction to PEG.

Summary: This study developed a questionnaire to investigate the eating habits of 109 women during pregnancy and statistically correlated the results with the data obtained by the newborn screening laboratory. The results showed that maternal lifestyle, diet, and drug intake during pregnancy may affect the neonatal metabolic profile, potentially leading to false positive or false negative results in the newborn screening test. The findings highlight the importance of understanding maternal nutrition and lifestyle in preventing misinterpretations of the neonatal metabolic profile, reducing unnecessary stress for newborns and their parents, and limiting costs for the healthcare system.

Development of a Method for Detection of SARS-CoV-2 Nucleocapsid Antibodies on Dried Blood Spot by DELFIA Immunoassay

Summary: The aim of this study was to improve the detection of SARS-CoV-2 nucleocapsid antibodies by developing a simple and reliable DELFIA immunoassay on dried blood spots (DBSs). The results showed that the DBS-DELFIA had a wider dynamic range and higher sensitivity for detecting SARS-CoV-2 nucleocapsid antibodies. Furthermore, a strong correlation was found between the DBS-DELFIA and ELISA immunoassays. Therefore, the combination of dried blood sampling with DELFIA technology provides an easier, minimally invasive, and accurate measurement of SARS-CoV-2 nucleocapsid antibodies in previously infected individuals.
