Lipoprotein-associated phospholipase A2 (LP-PLA2) may play an important role in the pathophysiology of coronary heart disease (CHD). The polymorphism of LP-PLA2 gene caused LP-PLA2 enzyme activity depressing or lost. But there is not a definite conclusion for the association of between the LP-PLA2 gene polymorphism and CHD risk. To assess the relationship between LP-PLA2 gene V279F polymorphism and CHD, a comprehensive Meta-analysis was performed. All the case-control studies evaluating the association of between the LP-PLA2 gene V279F polymorphism and CHD risk were identified. Seven case-control studies involving 3,614 patients with CHD and 4,334 controls were included. The crude odds ratios (ORs) of meta-analysis under the different gene model were not significant. But in the stratified analysis by study size, ethnicity, cases definition, and source of controls under the additive model, the association was evident in ethnicity for Japanese group (OR = 1.38, 95%CI = 1.22-1.56), cases definition for MI (OR = 1.22, 95%CI = 1.01-1.49), source of controls for the based-hospital (OR = 1.42, 95%CI = 1.24-1.59). These data suggested that the V279F polymorphism in LP-PLA2 gene may contribute to CHD development. But there is necessary that more well-designed large studies are required for the validation of this association.

Examining positional difference in basketball players' field goal accuracy using Bayesian Hierarchical Model

Feng Wang, Guohua Zheng

Summary: This study developed a Bayesian Hierarchical Modeling Scheme to analyze the positional difference of field goal accuracy at different shooting distances. The results showed that the overall accuracy decreases as the shooting distance increases. Different positions have varying shooting accuracy, and players shooting from over 24 feet have improved their field goal accuracy in recent years.

Celecoxib attenuates hepatosteatosis by impairing de novo lipogenesis via Akt-dependent lipogenic pathway

Cong Zhang, Yuzhen Lu, Yingying Song, Liang Chen, Junjie Hu, Yan Meng, Xin Chen, Shan Li, Guohua Zheng, Zhenpeng Qiu

Summary: Mounting evidence suggests that hepatic de novo lipogenesis is common in patients with non-alcoholic fatty liver disease (NAFLD). This study investigates the potential of the selective COX-2 inhibitor, celecoxib, to alleviate hepatic steatosis by targeting an Akt-driven lipogenic pathway. The results show that celecoxib effectively suppresses Akt/mTORC1 signaling and its downstream lipogenic cascade, thereby ameliorating hepatic steatosis in Akt-triggered NAFLD mice. However, its efficacy is limited in fructose-fed NAFLD mice.

Urolithin A Attenuates Hyperuricemic Nephropathy in Fructose-Fed Mice by Impairing STING-NLRP3 Axis-Mediated Inflammatory Response via Restoration of Parkin-Dependent Mitophagy

Cong Zhang, Yingying Song, Liang Chen, Peng Chen, Ming Yuan, Yan Meng, Qi Wang, Guohua Zheng, Zhenpeng Qiu

Summary: Urolithin A (UroA) has shown potential in alleviating fructose-induced hyperuricemic nephropathy by promoting Parkin-dependent mitophagy and suppressing STING-NLRP3 axis-mediated inflammatory response. It attenuated kidney damage and reduced inflammation in mice and human kidney cells, making it a promising intervention for preventing chronic kidney disease progression.

Summary: Acetylcholinesterase (AChE) inhibition is a common treatment for early-stage dementia and neurodegenerative diseases. A method using ultra-high-performance liquid chromatography-diode array detector coupled with AChE biochemical detection was established to rapidly identify AChE inhibitors (AChEIs) in Coptidis Rhizoma (CR) and evaluate its quality. The study identified several AChEIs in CR and highlighted their potential for neurodegenerative prevention and treatment.

The development and evaluation of hyaluronic acid coated mitochondrial targeting liposomes for celastrol delivery

Simeng Xiao, Siying Huang, Xiaojing Yang, Yujie Lei, Mingxiang Chang, Junjie Hu, Yan Meng, Guohua Zheng, Xinyan Chen

Summary: A novel multistage-targeted celastrol delivery system (C-TL/HA) was developed by electrostatic binding of hyaluronic acid (HA) to celastrol-loaded cationic liposomes. The study showed that C-TL/HA successfully delivered celastrol into mitochondria, activated apoptosis of mitochondrial pathway, and exhibited higher tumor inhibition efficiency and lower toxic side effects compared with free celastrol. The HA coating not only improved stability and safety in vivo, but also enhanced drug uptake and tumor targeting through recognition of CD44 receptors.

Radical Arylaminoformylation of Activated Alkenes to Amides Containing All-Carbon Quaternary Stereocenters

Summary: Here, we present an Ag+/S2O82--induced method for the synthesis of functionalized succinyl diamides with an all-carbon quaternary stereocenter via a radical aminoformylation/aryl-migration/desulfonylation cascade. This improved process utilizes oxalic monoamide, which is readily available and inexpensive, as a carbamoyl radical precursor through radical Smiles rearrangement. Additionally, the practical value of this synthesis strategy is enhanced by its tolerance to oxygen and water, operational simplicity, convenient reagents, and scalability.

Gallic acid impairs fructose-driven de novo lipogenesis and ameliorates hepatic steatosis via AMPK-dependent suppression of SREBP-1/ACC/FASN cascade

Yuzhen Lu, Cong Zhang, Yingying Song, Liang Chen, Xin Chen, Guohua Zheng, Yuan Yang, Peng Cao, Zhenpeng Qiu

Summary: Accumulated evidence suggests that de novo lipogenesis is a characteristic of NAFLD progression. Gallic acid (GA) is a phenolic acid with clinical significance in metabolic diseases. This study evaluated the potential of GA in treating fructose-induced NAFLD in mice. GA alleviated hepatic steatosis, oxidative stress, and inflammatory response in fructose-fed mice by restoring AMPK alpha phosphorylation and suppressing pro-lipogenic factors. The study demonstrates that GA supplementation may be a promising therapeutic strategy in NAFLD, especially in cases with enhanced hepatic lipogenesis.

Ellagitannins-Derived Intestinal Microbial Metabolite Urolithin A Ameliorates Fructose-Driven Hepatosteatosis by Suppressing Hepatic Lipid Metabolic Reprogramming and Inducing Lipophagy

Cong Zhang, Yingying Song, Ming Yuan, Liang Chen, Qianyu Zhang, Junjie Hu, Yan Meng, Shan Li, Guohua Zheng, Zhenpeng Qiu

Summary: This study investigated the efficacy of urolithin A (UroA) in a fructose-induced NAFLD mouse model and found that UroA alleviated hepatic steatosis. Mechanistically, UroA impaired lipogenesis and enhanced fi-oxidation in the livers of fructose-fed mice by facilitating hepatic lipophagy through the AMPK/ULK1 pathway. These findings suggest that dietary supplementation of UroA or ellagitannins-rich foods may be beneficial for NAFLD individuals with high fructose intake.

Summary: Multidrug resistance is a major challenge in chemotherapy for non-small cell lung cancer. Mitochondria have been identified as a promising target for overcoming drug resistance. A two-stage targeted liposome was successfully developed, with paclitaxel-loaded cationic liposome formulated and further coated with hyaluronic acid. The encapsulated paclitaxel showed improved stability and safety, and the liposomes enhanced cellular uptake and accumulation of paclitaxel in mitochondria, leading to disrupted mitochondrial function, increased apoptosis, and stronger anticancer efficacy.
