Summary: It has been established that T cells play a significant role in anti-tumor immune response, while the contribution of B cells has been insufficiently studied. In this research, the proportions and characteristics of B cells in tumor draining lymph nodes (TDLNs) and non-TDLNs from oral squamous cell carcinoma patients were analyzed. The results showed that TDLNs had a higher proportion of B cells, especially immunosuppressive regulatory B cells, compared to non-TDLNs. The presence of regulatory B cells in TDLNs may hinder the effectiveness of novel cancer immunotherapies in head and neck cancer.

Summary: The intense infiltration of regulatory T cells (Tregs) facilitates the qualities of glioma stem cells (GSCs) through TGF-beta secretion and NF-κB-IL6-STAT3 signaling pathway, leading to increased cancer stemness and tumorigenic potential. Blocking the IL6 receptor can mitigate the promotion effect of Tregs on glioma growth, and the expression levels of CD133, IL6, and TGF-beta could serve as prognosis markers for glioma patients.

Summary: Activation of B cells expressing CD25 can be enhanced by low-dose recombinant IL-2, leading to increased secretion of IL-10. Administration of low dose IL-2 to patients results in an increase in IL-10-producing B cells. Single-cell RNA sequencing reveals enrichment of regulatory B cell gene signature in plasma cell populations from IL-2-treated patients, with significant down-regulation of the transcriptional repressor BACH2. BACH2 suppression leads to increased B cell IL-10, suggesting its role in promoting an immunoregulatory phenotype in B cells through IL-2 modulation.

Summary: In this study, we successfully invalidated the ALDH1L2 gene using CRISPR-cas9 technique and found that the knockout of this gene rendered U251 glioblastoma cells methionine-independent in terms of cancer stem cell growth. We also observed higher levels of reactive oxygen radicals (ROS) and reduced total cellular NADPH in the knockout cells. The slower growth of the knockout tumor sphere may be related to its partial differentiation.

Summary: Mesenchymal stem cells (MSCs) play crucial roles in the tumor microenvironment (TME), contributing to tumor progression and therapeutic resistance. Glioma-resident MSCs (GR-MSCs) enhance the tumorigenicity of glioma stem cells (GSCs) through the IL-6/gp130/STAT3 pathway. Higher percentage of GR-MSCs in the TME leads to poor prognosis of glioma patients and their secretion of specific miRNA promotes tumor growth. Novel therapeutic strategies targeting GR-MSCs are urgently needed for GBM patients.

Summary: In cancer treatment, mesenchymal stem cells (MSCs) exhibit tropism towards colon cancer stem cells (CCSCs), particularly CD133(+)/CD44(+) cells. When recruited by CCSCs, BMSCs transform into cancer-associated fibroblasts and secrete more IL-8, resulting in increased stemness of CCSCs. Targeting the IL-8/CXCR2 pathway in BMSCs could potentially improve therapy targeting CCSCs.
