The expression and activation of ERK/MAPK pathway in human esophageal cancer cell line EC9706

While there have been more and more studies concerning mitogen-activated protein kinases (MAPKs) signaling pathways, which control many cellular complex programmes, such as cell proliferation, differentiation, cell death and embryogenesis. However, few studies are carried out about expression and activation of classical MAPKs, extracellular signal-regulated kinase1/2 (ERK1/2) in human esophageal cancer cell line. Therefore, in the present study, we investigated the expression and activation of ERK1/2 in human esophageal cancer cell line EC9706 and human normal esophageal epithelial cell line Heepic, which is as control. This study showed that ERK1/2 was transiently phosphorylated both in EC9706 and Heepic, the kinetics of which were slightly different. To further study the ERK/MAPK signaling pathway in EC9706 and Heepic cell line, U0126 a kind of specific inhibitor of MEK was used. This study showed that U0126 can block the phosphorylation of ERK1/2 in a short time, the complete inhibition concentration for EC9706 and Heepic cell line is 50 and 20 mu M, respectively. Incidentally, to further investigate the different roles of ERK1 and ERK2, vector-based short hairpin interference vectors targeted on ERK1/2 was constructed. Moreover, the effective interference target sequence was screened out in a transient transfection manner. MTT experiment showed that ERK2 is more important than ERK1 in the proliferation of EC9706 cells.

Summary: This study investigated the distribution and significance of tumor-infiltrating lymphocytes in the acidic tumor microenvironment of esophageal squamous cell carcinoma (ESCC). The findings showed that the density of cells stained with MCT4 in the epithelium was significantly associated with overall survival. Dendritic cells stained with S100 in the epithelial compartment correlated with clinical stage and tumor invasion depth.

Summary: The purpose of this study was to investigate the molecular mechanisms underlying the involvement of LINC00858 in colon cancer. The study found that LINC00858 was upregulated in colon cancer tissues and cells and promoted colon cancer progression by recruiting RAD21 and activating the PCNP-mediated STAT3/5 pathway. This provides a new therapeutic target for colon cancer treatment.

Mechanistically, it was found that LINC02820 may affect cytoskeletal remodeling, interact with SF3B3, and cooperate with TNFα to amplify the NF-kappa B signaling pathway, leading to ESCC metastasis.

Summary: SLC1A5 expression is upregulated in various cancers, including ESCC, but its immunological involvement in the tumor microenvironment of ESCC has been poorly described. In this study, in silico analyses and experimental confirmation were performed to investigate the prognostic and clinicopathological significance of SLC1A5 expression in ESCC. The results showed that SLC1A5 expression was unrelated to prognosis but correlated with CD8+ T-cell exclusion.

Clinicopathological Features and Prognostic Evaluation of UBR5 in Liver Cancer Patients

Summary: This study found that UBR5 expression is significantly increased in liver cancer patients and is positively correlated with patient age, tumor grade, lymph node metastasis, and survival rate. Furthermore, UBR5 was found to promote liver cancer cell proliferation by affecting YWHAZ expression. Experimental evidence confirmed that patients with higher levels of YWHAZ expression in liver cancer have poorer outcomes. These findings suggest that UBR5 associated with YWHAZ may influence the prognosis of liver cancer patients, and UBR5 could be a potential target for liver cancer treatment.
