Summary: This article reviews the role of gamma-tubulin in microtubule nucleation and its potential functions. Recent studies have provided insights into the structure, activation, and centrosomal recruitment of the gamma-tubulin ring complex (gamma-TuRC), shedding light on the molecular mechanism of microtubule nucleation. Furthermore, gamma-tubulin may also have specific nuclear functions and be involved in cross-talk between microtubules and microfilaments at the centrosome.

Summary: Yang et al. uncover a mechanism by which the centrosomin motif 1 (CM1) regulates microtubule nucleation and cellular organization. This regulation is achieved through an autoinhibitory mechanism and can be relieved by phosphorylation. This study provides valuable insights into the control of microtubule formation in cells.

Summary: The study successfully reconstituted the human gamma-TuRC and generated a subcomplex lacking MZT1 and actin. Comparisons showed that the subcomplex without the lumenal bridge can nucleate microtubules similarly to the holocomplex but with structural differences.

Summary: In this study, we provide the first real-time observation that γ TuNA directly increases γ TuRC activity, confirming it as a bona fide γ TuRC activator. We further validate this effect and demonstrate that γ TuNA's activating effect opposes inhibitory regulation by stathmin.

Summary: Microtubules, essential cytoskeletal elements assembled from ab-tubulin dimers, have their nucleation initiated by the gamma-TuRC complex. Recent cryoelectron microscopy studies and in vitro assays have revealed the molecular organization and potential activation mechanisms of gamma-TuRC, deepening our understanding of microtubule nucleation mechanisms.

Summary: In this study, it was found that fission yeast undergoes premeiotic nuclear oscillation, which is dependent on microtubules and is driven by cytoplasmic dynein. The oscillation frequency is cell length-dependent and is controlled by a balance between microtubule and viscous drag forces, as well as microtubule dynamics. These findings provide a basis for understanding the physical properties of microtubule-dependent nuclear movements.

Summary: Researchers identified novel small molecules that inhibit microtubule organization through sequential phenotypic screening of fission yeast and human cells. In vitro assays showed that these compounds directly bind tubulin and destabilize the structures of tubulin dimers. One compound in particular, L1, demonstrated preferential potency against human breast cancer cell lines and synergistic effects when combined with the Plk1 inhibitor BI2536, suggesting it as a candidate for cancer chemotherapy.

Summary: In this study, new components of the subdistal appendage (SDA), alpha-taxilin and gamma-taxilin, were identified and found to form a complex and play a role in the hierarchical assembly of the SDA. Additionally, the molecular composition and functional understanding of the SDA were revealed.

Summary: Epigenetic inheritance of heterochromatin requires mechanisms independent of DNA sequence, RNAi coupling, or input from DNA sequence. A DNA element termed maintainer has been identified that is sufficient for the epigenetic inheritance of pre-existing histone H3 lysine 9 methylation and heterochromatin in Schizosaccharomyces pombe. This maintainer is a composite DNA element with binding sites for specific transcription factors and ORC, and can be converted to a silencer in cells with lower rates of H3K9me turnover.

Summary: Saccharomyces cerevisiae can assimilate iron from the siderophore ferrichrome produced by other microbes via the Arn1 transporter. Schizosaccharomyces pombe synthesizes and secretes ferrichrome (Fc) using the sib1(+) and sib2(+) genes. Deletion of these genes in S. pombe impairs the ability to support the growth of S. cerevisiae when Fc is the sole source of iron. Further analysis identified the sib3(+) gene, encoding an N-5-transacetylase required for Fc production in S. pombe.

Summary: This study reveals the role of the augmin complex in microtubule branching through a structural model. By using cryo-electron microscopy and negative stain electron microscopy, the location and microtubule binding site of the augmin complex were identified. Evolutionary analysis shows that the structure and function of augmin are highly conserved across eukaryotes. These findings provide new insights into the mechanism of microtubule branching.

Summary: Cyclins and CDKs play key roles in eukaryotic cell biology, coordinating physiological conditions with cell cycle progression. Meiosis is a specialized developmental program ensuring proper genetic propagation to the next generation, with meiosis-specific cyclins being widespread in evolution. Studies have shown that the meiosis-specific Crs1 cyclin in fission yeast exhibits diversified CDK functions, with roles in meiotic S-phase progression, chromosome architecture maintenance, and other key meiosis-specific events.

Summary: Conditions altering the yeast cell wall lead to the activation of an adaptive transcriptional response mainly governed by the cell wall integrity (CWI) mitogen-activated protein kinase (MAPK) pathway. Two high-throughput screenings were conducted to identify genes related to cell wall integrity and transcriptional program elicited by cell wall stress. Functional groups related to chromatin organization, Golgi vesicle transport, rRNA processing, and protein glycosylation were found to be highly representative. Additionally, genes associated with RNA metabolism, transcription/translation, protein degradation, and chromatin organization were highlighted.

Summary: The study demonstrates that the cooperative maintenance of centriole engagement by Cep57 and Cep57L1 during interphase is crucial for controlling centriole copy number and ensuring proper chromosome segregation. Loss of Cep57 and Cep57L1 leads to premature centriole disengagement and subsequent errors in chromosome segregation.
