Summary: Diabetic cardiomyopathy (DCM) is a heart dysfunction that occurs in patients with diabetes mellitus (DM). This study found that dual-specificity phosphatase 12 (DUSP12) plays an important role in DCM. Overexpression of DUSP12 worsens cardiac dysfunction and increases oxidative stress and apoptosis, while DUSP12 deficiency exacerbates these effects. The study also found that DUSP12 interacts with a specific signaling pathway.

Summary: The extract from mushroom Gymnopilus purpureosquamulosus HOil (G. pps) showed cytotoxic effects on leukemia, lymphoma, and multiple myeloma cell lines by inducing apoptosis. The mechanisms involve changes in cellular redox status, activation of ASK1-MEK-SAPK/JNK and ASK1-P38 MAPK pathways, and inhibition of prosurvival proteins. Antioxidants alleviated the cytotoxicity of G. pps, highlighting its potential therapeutic value for hematologic malignancies.

Summary: Cancers are characterized by aberrant gene expression that leads to dysregulated cell division. Inhibiting these gene products has become a rational approach in cancer therapy. The protein ASK1, encoded by the MAP3K5 gene, is often found at elevated levels in cancer and has emerged as a potential target for developing chemotherapeutics. This study employed molecular modelling approaches to identify potential ASK1 inhibitors from phytochemicals. Three hit compounds, ellagic acid, luteolin, and kaempferol, showed promising inhibitory potentials and had suitable properties for further exploration in in vitro and in vivo studies.

Summary: In this experimental study, the mechanism of TBBPA toxicity on Grass carp hepatocytes (L8824 cells) and the antagonistic impact of EUC on TBBPA were investigated. The results showed that TBBPA exposure induced oxidative stress and activated the ASK1/JNK signaling pathway, resulting in apoptosis. Additionally, EUC effectively protected against TBBPA-induced apoptosis and immune dysfunction by regulating the ROS/ASK1/JNK pathway.
