Summary: PTEN is a frequently mutated gene in malignancies and functions as a powerful tumor suppressor, playing a role in multiple and complex processes of tumorigenesis. A newly identified isoform, PTENε/PTEN5, has been found to suppress tumor invasion and metastasis by interacting with VASP and ACTR2 to regulate filopodia formation and cell motility. The discovery of PTENε/PTEN5 advances our understanding of the diverse functions of PTEN.

Summary: Long noncoding RNAs (lncRNAs) are noncoding RNAs with length greater than 200 nt, which have been extensively studied for their biological roles and mechanisms. Hypoxia is a proven microenvironmental factor that promotes solid tumor metastasis, with epithelial-mesenchymal transition (EMT) being one of the major mechanisms induced by hypoxia. Many lncRNAs induced by hypoxia have been shown to interact with protein/protein complex and chromatin/epigenetic factors, contributing to metastasis.

Summary: In this study, a hypoxia-induced long non-coding RNA (lncRNA) RP11-367G18.1 was identified to be overexpressed in clear cell renal cell carcinoma (ccRCC) tissues. RP11-367G18.1 interacted with histone acetyltransferase p300 and regulated lysine 16 acetylation on histone 4 (H4K16Ac), thereby promoting ccRCC metastasis and growth. RP11-367G18.1 was associated with poor prognosis and may serve as a therapeutic target for ccRCC.
