Wnt/β-catenin signalling in adrenal physiology and tumour development

Wnt/beta-catenin signalling plays essential roles during embryonic development and in adult tissue homeostasis. Canonical signalling through Wnt secreted ligands relies on the control of beta-catenin cytoplasmic accumulation and translocation to the nucleus. In this compartment, beta-catenin serves as a transcription coactivator for transcription factors such as Lef/Tcf or some nuclear receptors. Constitutive Wnt signalling resulting from inactivation of inhibitors of the pathway or from activating mutations in beta-catenin, triggers tumour development in a number of tissues. Analysis of patients' samples and genetically engineered mouse models has shown that Wnt signalling was involved in adrenal development and tumourigenesis. This review will summarise all these recent findings and will focus on some of the mechanisms that may lead to aberrant accumulation of beta-catenin in adrenocortical tumours. Summary: This study found that high expression of HOXB9 and other HOX factors is associated with poorer prognosis in ACC patients. HOXB9 can promote adrenal tumor progression in a sex-dependent manner, and tumor cells are dependent on HOX function for survival. These findings have identified HOX factors as potential drug targets for novel therapeutic approaches in ACC.

Long-term follow-up and predictors of recurrence of Cushing's disease

Summary: Transsphenoidal surgery is the first-line treatment for Cushing's disease, with a remission rate of 70%-80%. However, the long-term remission rate is lower due to recurrence. Postoperative corticotroph insufficiency and its duration have been found to be protective factors against recurrence. Long-term monitoring with clinical and hormonal investigations is advised after postoperative recovery of the hypothalamic-pituitary-adrenal axis.

Summary: Bilateral macronodular adrenocortical disease (BMAD) is characterized by the development of adrenal macronodules resulting in a pituitary-ACTH independent Cushing's syndrome. In this study, the pathological features of 35 BMAD patients were analyzed, and 4 histopathological subtypes were identified. This classification highlights the heterogeneous pathological characteristics of BMAD, which correlate with certain genetic alterations identified in patients.

Summary: Defects in trophoblastic EMT caused by dysregulation of circTNRC18's interaction with LIN28A play a vital role in the development of preeclampsia. LIN28A overexpression suppresses circTNRC18-mediated inhibition of trophoblast migration, invasion, and EMT, while LIN28A knockdown promotes them. Furthermore, circTNRC18 regulates the intracellular distribution of LIN28A and the expression of insulin-like growth factor II, affecting cell migration and invasion. Targeting the circTNRC18-LIN28A regulatory axis may provide a novel treatment approach for preeclampsia.
