Adrenocortical tumor (ACT), a rare tumor with a heterogeneous presentation, incompletely understood pathogenesis, and generally poor prognosis, occurs in 1-2 people per million and is even more uncommon in the pediatric population. Such rare cancers are a challenge to clinical practice. Exchange of experience, information, and data on rare cancers is lacking, and outcomes for these rare cancers could be improved through the establishment of an international registry. The establishment of the International Pediatric Adrenocortical Tumor Registry (IPACTR) in 1990 by the St. Jude Children's Research Hospital International Outreach Program offered a new opportunity to collect clinical and laboratory features, treatment practices, and outcome data for children with ACT, research this disease, and systematically investigate how to improve patient outcomes. These efforts will improve the availability of information for both patients and the medical community. (C) 2011 Elsevier Ireland Ltd. All rights reserved.

Summary: The TP53-R337H founder mutation is highly prevalent in southern Brazil and is associated with pediatric adrenocortical tumors, early-onset breast cancers, and LFS. A knockin mutant p53 mouse model has been generated to study the variability in tumor susceptibility caused by this mutation.

Summary: Germline TP53 variants at exon 4-intron 4 junctions are frequent in pediatric adrenocortical tumors (ACT) and rare in other pediatric tumors. These variants result in improper expression and loss of function, and are associated with a high incidence of pediatric ACTs not consistent with classic Li-Fraumeni syndrome.

Summary: The study identified potential mechanisms of TERT activation in Wilms tumor, including mutations in the TERT promoter, increased methylation of the promoter, and genomic copy number amplifications. Conversely, inactivating WT1 mutations were associated with low TERT RNA levels and telomerase activity. N-MYC overexpression led to increased TERT promoter activity and transcription.

Summary: This study assessed the joint effect of environmental factors and the inherited TP53 p.R337H mutation on the development of breast cancer in three subregions of Southern Brazil. The results revealed that individuals living in heavily polluted environments are more susceptible to breast cancer due to this genetic mutation. Screening, genetic testing, medical improvements, and healthier lifestyle practices can help reduce the impact of environmental contaminants.

Summary: This study examined the effects of maternal malnutrition on the transcriptomic landscape of the ventral prostate in rats. It found that changes in molecular pathways related to cellular development and tissue morphogenesis were associated with maternal malnutrition. Summary: Defects in trophoblastic EMT caused by dysregulation of circTNRC18's interaction with LIN28A play a vital role in the development of preeclampsia. LIN28A overexpression suppresses circTNRC18-mediated inhibition of trophoblast migration, invasion, and EMT, while LIN28A knockdown promotes them. Furthermore, circTNRC18 regulates the intracellular distribution of LIN28A and the expression of insulin-like growth factor II, affecting cell migration and invasion. Targeting the circTNRC18-LIN28A regulatory axis may provide a novel treatment approach for preeclampsia.
