Summary: Prostate cancer bone metastases are common and often lead to painful skeletal events, with the BMP and TGF beta signaling pathways showing distinct effects in lytic or blastic bone lesions. Inhibiting BMP with DMH1 treatment restricts colonization of cancer cells in bone and preserves bone health. This suggests that profiling bone lesions in metastatic prostate cancer can help identify therapeutic targets to improve treatment outcomes.

Summary: BMP9 induces periodic increases of Hes1 mRNA and protein expression in osteoblasts through an autocrine negative feedback mechanism, which is significantly inhibited by an ALK inhibitor and overexpression of inhibitory Smad7. The transcriptional activation of Hes1 by BMP9 in osteoblasts is dependent on two Smad-binding sites in the upstream region of the gene.

Summary: This study identifies bone morphogenetic protein-9 (BMP-9) as a crucial component in cardiac remodeling after myocardial infarction (MI). Loss of BMP-9 leads to decreased survival, increased risk of cardiac rupture, increased collagen abundance, and enhanced matrix metalloproteinase-9 activity for collagen degradation after MI.

Summary: This study investigated the covalent binding of recombinant human bone morphogenetic protein-2 and -7 in stem cells and its effects on cell adhesion, migration, and osteogenesis. The presence of both proteins enhanced cell adhesion but limited cell motility, while the presence of single proteins promoted cell migration activity. Additionally, the coexistence of the proteins significantly increased early osteogenic marker expression.

Summary: Sertoli cells play a crucial role in spermatogenesis by forming the blood-testis barrier (BTB) and creating a unique microenvironment. This study reveals that a high-fat diet impairs BTB integrity and decreases sperm quality. The long noncoding RNA (lncRNA) Tug1 is found to be involved in the disruption of BTB by regulating Ccl2 and tight junction (TJ)-related proteins. Mechanistically, Tug1 directly binds to EZH2 and regulates H3K27me3 occupancy in the Ccl2 promoter region.

Summary: This study found that expression of HAVcR-1 is decreased in patients with prostate cancer, and the levels of HAVcR-1 ectodomain in serum are associated with the severity of the disease. In cell experiments, overexpression of HAVcR-1 led to changes in cell behavior and was associated with dysregulation of junctional complexes.

Summary: This study systematically investigated the expression of TRPV1 in LUAD and its relationship with tumor recurrence and metastasis. The results showed that increased TRPV1 expression was associated with worse overall survival outcome and raised HIF1 alpha levels in LUAD patients. TRPV1 overexpression promoted cell proliferation and migration, and the photothermal effect of CuS-TRPV1 mAb inhibited tumor growth and metastasis.

Summary: Cancer is a major cause of death worldwide, and drug resistance is a significant obstacle in cancer treatment. Osteopontin (OPN) is a protein that is overexpressed in various types of tumors and is involved in multiple cellular processes related to drug resistance. Understanding the role of OPN in cancer drug resistance may have implications for improving cancer therapy by reducing drug resistance in sensitive tumors.

Summary: Prohibitin (PHB) is a tumor suppressor gene that has multiple molecular activities. Overexpression of PHB leads to cell cycle arrest in the G1/S phase and represses the androgen receptor (AR) in prostate cancer cells. PHB interacts and represses members of the E2F family, which may also be linked to AR, making the AR:PHB:E2F interaction axis highly complex. Knockdown of PHB using siRNA increased the growth and metastatic potential of LNCaP mouse xenografts. On the other hand, ectopic overexpression of PHB affected the expression of several hundred genes in LNCaP cells, including downregulated expression of WNT family members and cell adhesion pathways. Online data analysis showed decreased PHB expression in clinical cases of metastatic prostate cancer, and its expression was correlated with higher WNT expression in metastasis. Overexpression of PHB reduced cell migration, motility, invasion, and attachment in prostate cancer cells. Mechanistically, AR was found to be involved in the regulation of the WNT genes, and these WNTs were also regulated by the cell cycle. Therefore, the repressive effects of PHB on AR, E2F, and WNT expression suggest its importance in inhibiting prostate cancer metastasis.

Summary: This study investigated the effect of vascular endothelial growth factor B (VEGFB) on regulating insulin secretion in type 2 diabetes mellitus (T2DM) and its underlying mechanism. The results showed that knockout of VEGFB exacerbated the decrease in insulin secretion in T2DM mice, while overexpression of VEGFB improved glucose tolerance and insulin sensitivity. VEGFB activated the Ca2+/CaMK2 pathway via VEGFR1-PLC gamma and IP3R pathway to regulate insulin secretion in T2DM.

Summary: This study investigates the influence of different heat treatment atmospheres on the microstructural and mechanical behaviors of SiC-ZrC fibers. Experimental results show that the fibers' mechanical performance declines under vacuum and argon environments. Under vacuum conditions, the amorphous phase decomposes and crystallinity increases, but no clustering of ZrC phase is observed. In contrast, under argon conditions, the amorphous phase decomposes and clustering of ZrC particles occurs, resulting in inferior mechanical properties of the fibers.
