Increase of carboxylesterase activity in Fasciola hepatica recovered from triclabendazole treated sheep

In the present work, we evaluate in vivo the activity of carboxylesterase of Fasciola hepatica exposed to triclabendazole. We observed a statistically significant increase in enzyme activity at 24 and 48 h post treatment (P <0.01 and P <0.001, respectively). The zymogram of cytosolic fractions identified a protein of 170 kDa containing the carboxylesterase activity. The densitograms of the zymograms confirmed the phenomenon of enzyme induction under the experimental conditions of the assay. These results provide not only the understanding of the importance of this metabolic pathway in flukes but carboxylesterase would also be an enzyme that could participate more actively in the development of anthelmintic resistance at TCBZ. Summary: Despite advancements in modern medicine, plant-derived medicines continue to be widely used due to their minimal side effects, low cost, and biocompatibility. This study evaluated the biological potential of methanolic and chloroform extracts from the roots, leaves, seeds, and flowers of Saussurea lappa. The extracts exhibited antioxidant, antibacterial, and cytotoxic activities, indicating their potential therapeutic applications. Further studies are recommended to isolate and characterize the bioactive compounds present in these plant parts for drug development.

Summary: The current study evaluated the phytochemicals and their potentials in Oxalis pes-caprae. The plant exhibited rich phenolic, flavonoid, alkaloid, and saponin contents, as well as significant antioxidative, antibacterial, and cytotoxic activities. Molecular docking analysis revealed that certain components of Oxalis pes-caprae showed good affinity with the SARS CoV-2 protein.

A Single Nucleotide Polymorphism Translates into a Radical Amino Acid Substitution at the Ligand-Binding Site in Fasciola hepatica Carboxylesterase B

Summary: The anthelmintic resistance of Fasciola hepatica may be related to the catalytic activity of xenobiotic metabolizing enzymes. An inducible enzyme, carboxylesterase B (CestB), was found to have a single nucleotide polymorphism at position 643 resulting in an amino acid substitution at position 215. This mutation affects the enzyme's affinity to inhibitors and ester ligands. A field genotyping survey showed the distribution of different alleles in parasites from Argentina and Mexico, without any linkage to host species or geographic origin.

Summary: The acaricidal activity of Datura innoxia ethanolic plant extract against Rhipicephalus microplus was studied using the adult immersion test and larval packet test. In vitro experiments and molecular docking simulations showed that the plant extract had consistent acaricidal activity and strong interactions with the target protein. This study also established the inhibitory effect of the extract on oviposition and larval mortality, indicating its potential use in tick management.

Acaro-pathogenic microorganisms, such as Aspergillus flavus strain INIFAP-2021, show potential as biocontrol agents against R. microplus.

Single Amino Acid Polymorphisms in the Fasciola hepatica Carboxylesterase Type B Gene and Their Potential Role in Anthelmintic Resistance

Summary: The expression of the CestB gene in Fasciola hepatica is induced by exposure to the anthelmintic triclabendazole (TCBZ), resulting in increased enzyme activity. Variations in the gene sequence can lead to radical amino acid substitutions at the ligand binding site, potentially impacting the interaction and catalytic properties of the enzyme. This study identified novel CestB polymorphisms in TCBZ-resistant parasites and evaluated their effects on the binding of the metabolically oxidized form of TCBZ (TCBZSOX) using 3D modeling. The findings suggest that these polymorphisms create a binding pocket for TCBZSOX and may contribute to drug sequestration type of anthelmintic resistance. These results lay the groundwork for future development of a molecular diagnostic tool for identifying anthelmintic resistance in F. hepatica.

Summary: This study explored the acaricidal effect and inhibitory mechanisms of Camellia sinensis plant extracts on ticks and mites. The results showed that the ethanolic plant extracts of C. sinensis exhibited high efficacy against mites and different stages of ticks in in vitro tests. In silico molecular docking revealed strong binding interactions between ellagic acid and the SsGST protein, as well as between quercetin and the RmGST protein.

Transcriptome assembly dataset of anthelmintic response in Fasciola hepatica

Summary: Fasciola hepatica is a globally distributed zoonotic parasitic worm that has developed resistance to anthelmintic compounds, prompting scientific efforts to study the molecular basis of this resistance for improved treatments. Transcriptome sequencing of sensitive and resistant strains treated with Triclabendazole and Albendazole revealed numerous new genes and RNA transcripts, providing insights into the mechanisms of resistance in these parasites.
