Different series of fluorescent agonists and antagonists have been developed and characterized for argininevasopressin and oxytocin G protein-coupled receptors. Both cyclic and linear peptide analogs of the neurohypophysial hormones are useful tools for investigating receptor localization and trafficking, analysing receptor structural organization, and developing new receptor-selective high-throughput ligand screening assays.

Summary: This study identified 8 novel mambaquaretin-1 (MQ1) peptides with nanomolar affinities for the V2R, all of which act as antagonists. These MQs form a new subgroup in the Kunitz family, closely related to the non-active dendrotoxins and two V2R-active cobra toxins. Sequence comparison between active and non-active V2R Kunitz peptides revealed key positions involved in V2R interaction. The MQ1-K39A variant showed a higher affinity for the human V2R.

Molecular insights into mechanisms of GPCR hijacking by Staphylococcus aureus

Functional Rescue of a Nephrogenic Diabetes Insipidus Causing Mutation in the V2 Vasopressin Receptor by Specific Antagonist and Agonist Pharmacochaperones

Summary: In this study, a point mutation in the AVPR2 gene of a patient with nephrogenic diabetes insipidus (NDI) was identified, and the impaired function of the mutant receptor was characterized. The mutant receptor was primarily located in the endoplasmic reticulum (ER) and had reduced ability to generate cAMP in response to AVP stimulation. Pretreatment with pharmacochaperones partially restored the receptor's function. Both cell permeant agonists and antagonists can function as pharmacochaperones, providing a potential starting point for developing therapies for patients carrying this mutation.

Structure of the vasopressin hormone-V2 receptor-β-arrestin1 ternary complex

Summary: Recent structural studies have provided insights into the molecular mechanisms of GPCR-arrestin coupling, but it is unclear whether this process is conserved among different GPCRs. In this study, the cryo-electron microscopy active structure of the wild-type arginine-vasopressin V2 receptor (V2R) in complex with beta-arrestin1 was determined, revealing a unique arrangement of beta-arrestin1 and an interface involving all receptor intracellular loops. These findings suggest a significant structural variability among GPCR-arrestin signaling complexes.
