Captive maturation studies in Penaeus monodon by GIH silencing using constitutively expressed long hairpin RNA

Captive maturation and spawning in Penaeus monodon are currently induced by unilateral eyestalk ablation, a method that removes the gonad-inhibiting hormone (GIH) secreted from the eyestalk. However, unilateral eyestalk ablation creates physiological complications in the brooder due to the unintended removal of other hormones secreted by the eyestalks. Here we studied the effect of gih silencing by long hairpin RNA on the reproductive physiology of P. monodon. We observed 3-5 fold enhanced expression of the androgenic gland hormone (AGH) transcript in males, but no effect on vitellogenin expression in females. In the destalked animals, however, positive effect on the maturation indicator transcripts was seen in both the sexes, and surpassed the efficiency of the silencing treatment. There were indications of active spermatogenesis in histological sections of both the gih silenced and destalked males, but no changes at histological level in the females. We also observed a significantly higher rate of molting and associated mortality in the gih silenced animals at the end of the experiment. Our results suggest that eyestalk ablation is still the most efficient technique to induce maturation in P. monodon. Although gih silencing could be a potential alternative to eyestalk ablation, further research is needed to enhance its efficiency over eyestalk ablation by using tissue specific and/or inducible promoter for lhRNA expression. It is also evident that females need to be fairly large in size for these interventions to succeed. Statement of relevance Induction of captive maturation has been a challenge for shrimp industry. The seed production is often hampered by the low abundance and high price of naturally collected broodstock. Collection of natural brooders also has the intrinsic problem of WSSV infection. Also, genetic improvement of this highly preferred species is difficult unless the reproductive cycle is closed. For all these reasons it is important to study the reproductive physiology and endocrinology of this species and ways to manipulate it in order to induce captive maturation. The present study utilizes the RNA interference mechanism to silence the gonad-inhibiting hormone gene to see its effect on maturation. (C) 2015 Elsevier B.V. All rights reserved.

Gain of function studies on predicted host receptors for white spot virus

Summary: Three decades after its first outbreak, the shrimp white spot virus (WSV) is still a global cause of concern. The pathogenesis of WSV is not clearly understood, despite the identification of several candidate host receptor proteins. In this study, protein-protein docking was used to evaluate the interaction between the WSV envelope protein VP28 and four host receptors. Furthermore, gain-of-function experiments confirmed that none of the receptors alone were sufficient to make cells permissive to WSV, suggesting the involvement of co-receptors.

Summary: The present study characterized the complete mitochondrial genome of the Blue-spotted maskray, Neotrygon indica, and analyzed its evolutionary relationship within the Dasyatidae family. The study found distinct characteristics of N. indica through comparative mitogenomics and revealed variations in codon usage among different species of the family. The phylogenetic tree indicated a sister relationship between the Dasyatinae and Neotrygoninae subfamilies, with N. indica forming a sister species to a clade consisting of N. varidens and N. orientalis.

Meta-analysis of fish stock identification in India: current status and future perspectives

Summary: A meta-analysis was conducted on fishstock identification research in India between 2000 and 2021. The study highlighted the need for strengthening stock identification studies and addressing the limitations in research, such as the lack of studies on marine species and elasmobranchs.

New Record of Rare Skeletal Anomaly, Pug Head Deformity in Striped Catfish, Mystus tengara (Hamilton, 1822) from India

Summary: A deformed specimen from a fish market in West Bengal, India was studied and compared with a normal specimen from the same location. The deformed specimen exhibited a shorter upper jaw, a blunt front part of the head, and a reduced snout length. No significant variation was observed in meristic characters. Possible causes for the deformity include chemical contaminants, dietary limitations or excesses, temperature variations, or genetic mutation during larval development. This is the first report of pug head deformity in Mystus tengara from India.
