A Comparison of Imaging Methodologies for 3D Tissue Engineering

Imaging of cells in two dimensions is routinely performed within cell biology and tissue engineering laboratories. When biology moves into three dimensions imaging becomes more challenging, especially when multiple cell types are used. This review compares imaging techniques used regularly in our laboratory in the culture of cells in both two and three dimensions. The techniques reviewed include phase contrast microscopy, fluorescent microscopy, confocal laser scanning microscopy, electron microscopy, and optical coherence tomography. Treatment of murine partial thickness scald injuries with multipotent adult progenitor cells decreases inflammation and promotes angiogenesis leading to improved burn injury repair

Summary: MAPC cells have shown promising effects on burn injury repair by inhibiting T cell proliferation, reducing inflammation, and promoting angiogenesis. When injected into burn wounds, MAPC cells can decrease wound area, width, and increase re-epithelialization, as well as dampen inflammation and improve angiogenesis in the burn area. However, extracellular matrix production was not significantly affected by MAPC cell treatment. These findings suggest that MAPC cells could be a potential candidate for clinical cell therapies in burn injury repair.

Mechanistic Insight in Surface Nanotopography Driven Cellular Migration

Summary: The study showed that surfaces with larger nanoscale topography lead to faster closure of gaps between cell monolayers, mainly driven by cell migration independent of cell proliferation. Surface nanotopography significantly modulates the signaling pathways and proteins involved in cell migration by directly recruiting receptor and adapter proteins responsible for cell-surface interaction.

Summary: Advances in cellular reprogramming have improved the use of patient-derived cells for neurological research. However, the adherence of human neurons on tissue cultureware is unreliable, hindering their electrophysiological maturation. Thin-film plasma polymer treatments and extracellular matrix coatings can enhance the adherence of human neuronal cultures on glass and improve their electrophysiological properties.
