Listeria monocytogenes (Lm) invades the host intestine using listerial invasion proteins, internalins. The in vivo role of internalin A (InlA) and internalin B (InlB) is reported here. Intragastric (i.g.) administration and ligated loop assays with delta inlB-Lm demonstrated that a lack of InlB significantly attenuates the invasive ability of Lm into various organs. On the other hand, InlAm-Lm expressing a mutant InlA with two substitutions, S192N and Y369S, which has been reported to increase the affinity of InlA to mouse E-cadherin, resulted in little increase in intestinal infection according to both ligated loop and i.g. infection assays. Lm preferentially enters ileal Peyer's patch (PP) via M cells and delta inlB-Lm showed severely reduced ability to invade though these cells. The present results reveal the importance of InlB, which accelerates listerial invasion into M cells on ileal PPs in vivo.

Summary: The study revealed that metabolites produced by Helicobacter pylori interact with host cholesterol and exacerbate gastritis through C-type lectin receptors. The findings from mutant H. pylori strains lacking specific metabolites showed impaired ability to cause gastritis.

Innate Lymphoid Cells in Skin Homeostasis and Malignancy

Marek Wagner, Shigeo Koyasu

Summary: Innate lymphoid cells (ILCs), enriched in barrier tissues like skin, play a role in immune pathways despite lacking antigen receptors found on T and B cells. They are crucial in controlling melanoma and potentially benefiting patients with skin malignancies through their therapeutic potential.

ILC2s and Adipose Tissue Homeostasis: Progress to Date and the Road Ahead

Takuma Misawa, Marek Wagner, Shigeo Koyasu

Summary: ILC2s are tissue-resident immune cells that exist not only in adipose tissue but also in various other tissues such as lung and skin. They can change their functions or distribution depending on the environment and circumstances. This review outlines the recent understanding of ILC2 biology and discusses future directions for ILC2 research, particularly in adipose tissue and metabolic homeostasis.

Summary: Insulin resistance is the primary pathophysiology underlying metabolic syndrome and type 2 diabetes. Carbohydrate metabolism by gut microbiota plays a crucial role in insulin resistance, and an increase in host-accessible monosaccharides is associated with insulin resistance and host inflammation.

Summary: This study reveals the silent spread of New Delhi metallo-beta-lactamase (NDM) carbapenemase in Brazil, particularly in Enterobacterales other than Klebsiella spp. It highlights the diverse species and coexistence of important resistance genes in these NDM-producing strains. Prompt detection and effective measures are crucial to mitigate the further dissemination of NDM carbapenemase and preserve antimicrobial therapy options in hospital settings.
