Optimized 25-hydroxyvitamin D analysis using liquid-liquid extraction with 2D separation with LC/MS/MS detection, provides superior precision compared to conventional assays

The analysis of 25-hydroxyvitamin D3 (25(OH)D3) and related metabolites represents a considerable challenge for both clinical and research laboratories worldwide. There is currently debate about the best methodology employed to assess vitamin D status and whether the 3-epi-25-hydroxyvitamin D3 (3-epi-25(OH)D3) should be separated and quantitated when measuring 25(OH)D3. Mass spectrometry techniques are generally regarded as the gold standard due to high specificity for vitamin D metabolites. However, many methods require high sample volumes for analysis. We have developed a new 2 dimensional (2D) ultra performance liquid chromatography (UPLC) separation coupled tandem mass spectrometry (MS/MS) detection to accurately quantitate 25(OH)D3, epi-25(OH)D3, and 25(OH)D2 in adults and children, requiring only 50 mu L of human serum. The assay gives excellent separation of epi-25(OH)D3, and 25(OH)D2 from 25(OH)D3, has excellent precision with an intra-assay CV of 0.5 % at 74 nmol/L and can report down to 2 nmol/L for 25(OH)D3. Furthermore, the method shows excellent agreement with the vitamin D external quality assessment scheme (DEQAS) quality control program for vitamin D analysis. We present this approach as a candidate reference method for 25(OH)D3, epi-25(OH)D3, and 25(OH)D2 analysis in humans.

Summary: The Vitamin D Standardization Program (VDSP) conducted an interlaboratory comparison study to evaluate the performance of liquid chromatography - tandem mass spectrometry (LC-MS/MS) assays for determining serum total 25-hydroxyvitamin D. Results showed that only 53% of the assays met the VDSP criterion, with four assays having significant performance issues due to the lack of separation between 3-epi-25(OH)D-3 and 25(OH)D-3.

Quantifying the effectiveness of betaherpesvirus-vectored transmissible vaccines

Summary: This study evaluates the effectiveness of using betaherpesviruses as transmissible vaccine vectors. The mathematical model developed and parameterized using data from mouse populations infected with murine cytomegalovirus (MCMV) shows rapid and effective control of pathogens with the introduction of the transmissible vaccine. However, the effectiveness may vary across reservoir populations and specific vector strains used for the vaccine.

Summary: Antenatal steroids play an important role in promoting functional maturation of the fetal lung. Current dosing regimens may pose potential risks, but this study found that direct fetal administration of a low dose of betamethasone phosphate can achieve the same lung maturation without maternal steroid administration.

Summary: This study established new routes for the chemical synthesis of vitamin D-3 and its hydroxy metabolites. The compounds showed similar properties in inhibiting cell proliferation and inducing cell differentiation, as well as effects on T-cell immune activity. They also interacted with nuclear receptors in a similar manner. The findings have important implications for studying the role of these compounds in the immune system and inflammatory diseases.

Summary: This study used molecular simulation to investigate the interaction between vitamin D3 hydroxyderivatives and AhR. It was found that these derivatives can stimulate downstream target genes of AhR. Molecular dynamics simulations showed favorable binding of the derivatives to AhR and revealed potential mechanisms underlying their binding affinities.

Summary: CYP11A1 and CYP27A1 hydroxylate tachysterol(3), a photoproduct of previtamin D-3, producing 20S-hydroxytachysterol(3) and 25(OH)T-3, which are biologically active and interact with VDR, AhR, LXRs, and PPAR gamma. These metabolites inhibit cell proliferation and stimulate differentiation and anti-oxidative gene expression in keratinocytes and fibroblasts.

Analysis of the ability of vitamin D3-metabolizing cytochromes P450 to act on vitamin D3 sulfate and 25-hydroxyvitamin D3 3-sulfate

Summary: Our study found that 25-hydroxyvitamin D3 (25(OH)D3) can exist in the human circulation in the sulfated form, and can be metabolized by vitamin D-metabolizing cytochromes P450 (CYPs). Specifically, CYP27A1 can convert vitamin D3 sulfate to 25(OH)D3 3-sulfate, while CYP11A1 can convert vitamin D3 sulfate to 20(OH)D3 3-sulfate, but at a lower rate.

Summary: Vitamin D exists in two forms, D3 produced in the skin and D2 obtained from the diet. The enzyme CYP24A1 plays a crucial role in oxidizing and inactivating both forms of vitamin D, regulating their serum levels. This study aimed to clarify the pathways of oxidation for 25(OH)D2 and 1,25(OH)(2)D2 and compare their metabolism with their vitamin D3 counterparts.

Metabolism of Lumisterol2 by CYP27A1

Summary: Lumisterol2 (L2) is a photoproduct of UVB action on ergosterol, which is present in edible mushrooms and human skin. CYP27A1 efficiently metabolizes L2 to produce three major hydroxy-L2 products, while CYP11A1 has minimal effect on L2 metabolism. The major CYP27A1-derived hydroxy-L2 products have anti-proliferative activity on skin-cancer cell lines.
