Summary: The ethanol extract of P. brevitarsis larvae (PBE) can inhibit RANKL-induced osteoclast formation and prevent bone loss. PBE decreases tartrate-resistant acid phosphatase activity and the expression of osteoclastogenesis-associated genes and proteins. It also inhibits the phosphorylation of p38 and NF-κB.

Summary: This study found that the expression of CGRP in the femur of osteoporotic patients was downregulated, while the expression of CGRP and serotonin in the spinal cord and brainstem was increased. These results suggest that the activation of osteoclasts may trigger the release of CGRP from sensory nerve fibers and promote serotonin release in the spinal cord and brainstem, contributing to bone loss in postmenopausal osteoporosis.

Summary: This study investigated the effects of periodontitis on osteoporosis. The results showed that the salivary microbiota of patients with periodontitis aggravated femoral bone resorption in rats with ovariectomy-induced osteoporosis. Additionally, intestinal inflammation was exacerbated after the administration of periodontitis salivary microbiota in these rats. The correlation analysis of microbiota and metabolomics revealed potential involvement of lipolysis and tryptophan metabolism in bone loss induced by periodontitis salivary microbiota.

Summary: Postmenopausal osteoporosis is a common metabolic disease in old women. Estrogen supplementation through bioactive substances, such as soybean isoflavone aglycones, has been studied for improving menopausal syndrome. This study evaluated the effect of different doses of high-purity soybean isoflavone aglycones on a rat model of osteoporosis. The results showed that the AFDP-H group had a similar intervention effect as the EE group and showed improvement in bone microstructure and prevented weight gain and increased cholesterol levels.

Summary: A novel nanocombination of flavonoids encapsulated in PLGA nanoparticles showed promising results for anti-osteoporotic therapeutics, demonstrating inhibitory effects on trabecular bone loss and significant improvements in bone mineral density and microarchitecture. The cytocompatibility of the nanoparticles was also confirmed in vitro. Further research may lead to novel therapeutic management of osteoporosis.

Summary: This study investigated the difference in the protective effects of intraperitoneal injection of exogenous melatonin during daytime or nighttime on bone loss in ovariectomized rats. Results showed that daytime treatment significantly increased bone mass in ovariectomized rats compared to nighttime treatment. Microscopic and biomechanical experiments revealed improved bone parameters and strength in the daytime treatment group. Molecular biology experiments showed increased bone formation-related molecules and decreased bone resorption-related molecules. Cell experiments demonstrated higher cell viability and better inhibition of ROS production and apoptosis with low-dose melatonin treatment.

Summary: Increased bone resorption caused by excessive osteoclasts is the main cause of osteoporosis. The expression of RILP, a protein, is induced by receptor activator of NF-KB ligand, and its inhibition decreases the number and function of osteoclasts. Inhibition of RILP also reduces preosteoclast migration and bone resorption. The study suggests that RILP plays a crucial role in osteoclast formation and bone resorption, and could be a therapeutic target for bone diseases caused by excessive osteoclasts.
