Summary: The levonorgestrel intrauterine device shows substantial activity in treating complex atypical hyperplasia and grade 1 endometrioid endometrial cancer, with a modest proportion of patients demonstrating upfront progesterone resistance. Potential biomarkers were identified that may correlate with resistance to therapy. Further exploration is warranted.

Summary: Women with complex atypical hyperplasia or early-stage endometrioid endometrial cancer can preserve fertility through progesterone therapy and deferral of hysterectomy. Loss of MIG-6 and PGR expression, as well as activation of pAKT, may lead to progesterone resistance in these conditions.

Summary: The adjuvant efficacy of metformin in achieving pathological complete response in patients with endometrial complex hyperplasia and complex atypical hyperplasia was investigated. Regression of CH/CAH may be improved by progesterone+metformin, especially in patients with BMI ≥ 25 kg/m(2) and patients with PCOS. Metformin had no significant effect on subsequent ART outcomes.

Summary: The study aimed to explore the health benefits of boosting NAD levels with NR on HCC. The results showed that NR supplementation alleviated HCC-induced weight loss and metastasis to the lung, and decreased HCC progression and metastasis in both in vivo and in vitro experiments. Therefore, supplementing NR may serve as an effective treatment for the suppression of HCC progression.

Summary: This study utilized massively parallel sequencing to detect cancer-associated mutations in endometrial lavage specimens. It found that endometrial hyperplasia, especially with atypia, is characterized by a high burden of cancer-associated mutations. The concordance rates between mutations identified in endometrial lavage specimens and endometrial biopsies were relatively low, indicating the potential of missed focal atypia. The study suggests that massively parallel sequencing of endometrial lavage is feasible and may allow tailoring of endometrial biopsies for individual risk assessment of atypical endometrial hyperplasia.

Summary: The transcription factor MGA, a member of the MYC network, plays a tumor suppressive role in vivo, as shown in mouse models of non-small cell lung cancer. Its loss accelerates tumor growth, enhances invasive capabilities, and affects the expression of genes associated with metastasis. MGA-MAX, E2F6, and L3MBTL2 co-occupy thousands of promoters, with MGA stabilizing these subunits for function.

Summary: This study evaluated the diagnostic value of shear wave elastography (SWE) in endometrial cancer (EC) and atypical endometrial hyperplasia (AEH), and found that combining SWE with conventional ultrasonography could improve diagnostic effectiveness. Logistic regression analysis identified endometrial thickness, Color score, and Young's modulus as independent risk factors for EC and AEH.

Summary: Both IR and MetS are significantly associated with recurrence and longer time to complete remission in young women with atypical endometrial hyperplasia (AEH) and early endometrial cancer (EC) undergoing fertility-sparing treatment. Using IR and MetS as markers can significantly improve the diagnostic accuracy of recurrence for patients undergoing fertility-sparing treatment and may have synergistic roles in suppressing treatment.

Summary: Genetic risk assessment for aggressive prostate cancer is challenging. In this study, a new polygenic risk score based on SNPs associated with prostate volume was developed and found to be able to predict the risk and mortality of aggressive prostate cancer. These findings provide a new genetic risk assessment method for the disease prognosis of prostate cancer patients.

Summary: This qualitative study aimed to understand the acceptability of bariatric surgery in young individuals with obesity and endometrial cancer or atypical hyperplasia. The study found that participants were reluctant to accept bariatric surgery due to lack of knowledge, stigma, and fear of risks associated with the procedure. There was a significant gap in participant understanding of the complex interplay between their cancer, infertility, and obesity. More support and patient-oriented counseling are needed before presenting bariatric surgery as a treatment option.

Weight loss of more than 10% has a positive influence on treatment response, recurrence, and pregnancy rates. GnRHa could be a suitable option for obese women.
