Transcriptomic Profiling and Gene Disruption Revealed that Two Genes Related to PUFAs/DHA Biosynthesis May be Essential for Cell Growth of Aurantiochytrium sp.

Aurantiochytrium sp. PKU#SW7 is a thraustochytrid strain that was found to exhibit high potential for docosahexaenoic acid (DHA, C22:6n-3) production. In this work, the transcriptome of Aurantiochytrium sp. PKU#SW7 was analyzed for the study of genes involved in basic metabolic functions and especially in the mechanisms of DHA biosynthesis. Sequence annotation and functional analysis revealed that the strain contains components of fatty acid synthesis (FAS) and polyketide synthase (PKS) pathways. Fatty acid desaturases and elongases were identified as components of FAS pathway, whilst key components of PKS pathway were also found in the cDNA library. The relative contribution of the two pathways to the synthesis of DHA was unknown, as both pathways appeared to be lacking full complement of genes for standalone synthesis of DHA. Further analysis of two putative genes encoding the very-long-chain (3R)-3-hydroxyacyl-CoA dehydratase and dehydrase/isomerase involved in FAS and PKS pathways, respectively, revealed that under various salinity conditions, their relative expression levels changed corresponding to the variation of DHA content in Aurantiochytrium sp. Independent knock outs of these genes in Aurantiochytrium sp. resulted in poor cell growth, probably due to little or no intracellular DHA accumulation. Hence, it can be speculated that both genes are engaged in DHA biosynthesis and DHA in Aurantiochytrium sp. could be produced by jointed actions of both FAS and PKS systems.

Effects of Sulfamethazine and Cupric Ion on Treatment of Anaerobically Digested Swine Wastewater with Growing Duckweed

Summary: This study investigated the effects of sulfamethazine (SMZ) and cupric ion on the removal of NH4+-N and total phosphorus (TP) from anaerobically digested swine wastewater (ADSW) by duckweed. Duckweed showed excellent nutrient removal efficiency and tolerance to SMZ stress. However, the combined application of SMZ and cupric ion inhibited nutrient removal while increasing the contents of photosynthetic pigments, proteins, and vitamin E. Potential degradation pathways of SMZ in the duckweed system were proposed. This research provides a novel approach for nutrient removal and value-added product production using the duckweed system under the stress of SMZ and cupric ion.

Characterization of a Novel Hot-Spring Cyanobacterium Leptodesmis sichuanensis sp. Nov. and Genomic Insights of Molecular Adaptations Into Its Habitat

Summary: The study describes several strains of filamentous cyanobacteria from different, primarily cold environments, belonging to the genus Leptodesmis. The complete genome of a novel hot-spring strain, Leptodesmis sp. PKUAC-SCTA121 (A121), isolated from Erdaoqiao hot springs in China, was sequenced. The results confirm the establishment of a new species within Leptodesmis, named Leptodesmis sichuanensis sp. nov. The study also provides the first insight into the genomic features of the genus Leptodesmis and lays the foundation for future global ecogenomic and geogenomic studies.

Efficient degradation of tetracycline by singlet oxygen-dominated peroxymonosulfate activation with magnetic nitrogen-doped porous carbon

Summary: The study thoroughly examined the degradation of tetracycline (TC) in the constructed Co-N/C-PMS system, finding that the removal rate of TC reached 85.4% when Co-N/C and PMS were added simultaneously, with a higher degradation rate compared to using Co-N/C or PMS alone. The system was identified as a singlet oxygen-dominated nonradical reaction, providing a promising approach for eliminating TC in water treatment.

Mechanism study on the regulation of metabolite flux for producing promising bioactive substances in microalgae Desmodesmus sp.YT through salinity stress

Summary: Desmodesmus sp.YT, rich in bioactive substances, showed impressive tolerance to various environmental conditions. This study investigated the metabolic transformation influenced by salt stress in Desmodesmus sp.YT through analysis of biochemical compositions and comparative transcriptome. Results showed that normal treatment enhanced biomass and protein production, while salt stress stimulated cellulose and starch biosynthesis. Desmodesmus sp.YT cultured in freshwater could be used as feed additives, while microalgae grown in seawater had potential for biofuel production.

Generation of miniploid cells and improved natural transformation procedure for a model cyanobacterium Synechococcus elongatus PCC 7942

Summary: This study optimized the cultivation and natural transformation protocol for the important cyanobacterium Synechococcus elongatus PCC 7942. By adjusting agar concentration and overexpressing catalase gene, the growth of Synechococcus 7942 on solid media was improved. High temperature and phosphate depletion conditions resulted in fewer genome copies and potential to produce fully segregated mutants. Higher antibiotic concentrations improved the selection of homozygous transformants.

FELICX: A robust nucleic acid detection method using flap endonuclease and CRISPR-Cas12

Chitosan-Fe3O4 composites enhance anaerobic digestion of liquor wastewater under acidic stress

Summary: In this study, chitosan-Fe3O4 was prepared and its effects on anaerobic digestion processes under acid stress were investigated. The results revealed that chitosan-Fe3O4 significantly increased the methanogenesis rate and accelerated the restoration of acidified anaerobic systems. The analysis of sludge characteristics indicated that chitosan-Fe3O4 promoted the secretion of proteins and humic substances, and enhanced the electron transfer activity of the system. Microbial community analysis showed that chitosan-Fe3O4 enriched the abundance of Peptoclostridium and facilitated direct interspecies electron transfer. These findings suggest that chitosan-Fe3O4 can be used to improve the efficiency of anaerobic digestion of high concentration organic wastewater under acid inhibition conditions.

Cupric ions inducing dynamic hormesis in duckweed systems for swine wastewater treatment: Quantification, modelling and mechanisms

Summary: Hormesis has been a topic of concern in the environmental and toxicological communities. Previous studies have focused on the hormesis induced by stressors in terms of their biotoxicity, but little research has been done on hormesis in the context of biological wastewater treatment processes. In this study, the removal of NH4+-N and Cu2+ by S. polyrrhiza was investigated under long-term exposure to environmentally relevant concentrations of Cu2+ in swine wastewater. The results showed that long-term exposure to Cu2+ enhanced the removal of NH4+-N by S. polyrrhiza, and previous exposure to low doses of Cu2+ significantly improved the removal efficiency in a second exposure.

Distinct Molecular Patterns of Two-Component Signal Transduction Systems in Thermophilic Cyanobacteria as Revealed by Genomic Identification

Summary: This study investigated the structure and architecture of two-component systems (TCSs) in 17 thermophilic cyanobacteria and revealed their complex and diverse nature. The results showed a unique composition of TCS genes in these cyanobacteria, which were possibly associated with various functions. Horizontal gene transfer and gene duplication events might have played a role in the evolutionary history of TCS genes in certain genera. These findings highlight the potential of thermophilic cyanobacteria genomes for adapting to environmental fluctuations.

Combined effects of oxytetracycline concentration and organic loading rate on semi-continuous anaerobic digestion of swine wastewater

Summary: The high concentrations of antibiotics in swine wastewater may have potential adverse effects on anaerobic digestion (AD). Previous studies focused on the effects of antibiotic concentrations but did not consider the fluctuation of swine wastewater quality and reactor operating conditions in practical engineering applications. This study found that continuous addition of oxytetracycline for 30 days had no effect on AD performance in systems with COD of 3300 mg/L and HRT of 4.4 days. However, when COD and HRT were changed to 4950 mg/L and 1.5 days respectively, oxytetracycline at different concentrations had varying effects on methane yield and cell membrane.
