Cellular Localization of Debromohymenialdisine and Hymenialdisine in the Marine Sponge Axinella sp Using a Newly Developed Cell Purification Protocol

Sponges (Porifera), as the best known source of bioactive marine natural products in metazoans, play a significant role in marine drug discovery and development. As sessile filter-feeding animals, a considerable portion of the sponge biomass can be made of endosymbiotic and associated microorganisms. Understanding the cellular origin of targeted bioactive compounds from sponges is therefore important not only for providing chemotaxonomic information but also for defining the bioactive production strategy in terms of sponge aquaculture, cell culture, or fermentation of associated bacteria. The two alkaloids debromohymenialdisine (DBH) and hymenialdisine (HD), which are cyclin-dependent kinase inhibitors with pharmacological activities for treating osteoarthritis and Alzheimer's disease, have been isolated from the sponge Axinella sp. In this study, the cellular localization of these two alkaloids was determined through the quantification of these alkaloids in different cell fractions by high-performance liquid chromatography (HPLC). First, using a differential centrifugation method, the dissociated cells were separated into different groups according to their sizes. The two bioactive alkaloids were mainly found in sponge cells obtained from low-speed centrifugation. Further cell purifications were accomplished by a newly developed multi-step protocol. Four enriched cell fractions (C1, C2, C3, and C4) were obtained and subjected to light and transmission electron microscopy, cytochemical staining, and HPLC quantification. Compared to the low concentrations in other cell fractions, DBH and HD accounted for 10.9% and 6.1%, respectively, of dry weight in the C1 fraction. Using the morphological characteristics and cytochemical staining results, cells in the C1 fraction were speculated to be spherulous cells. This result shows that DBH and HD in Axinella sp. are located in sponge cells and mostly stored in spherulous cells.

Marine Sponge Endosymbionts: Structural and Functional Specificity of the Microbiome within Euryspongia arenaria Cells

Summary: Sponges host abundant microbial symbionts that can produce an impressive number of novel bioactive metabolites. However, knowledge on intracellular (endosymbiotic) microbiota is scarce. By separating sponge cells and enriching intracellular microbes, we characterize the composition and function of the endosymbiotic microbiome, uncovering a noteworthy number of taxa exclusively in the endosymbiotic microbiome. Our study achieves a more comprehensive sponge microbial community profile, which demonstrates the structural and functional specificity of the endosymbiotic microbiome.

Summary: A shear-stress-mediated platform called portable vortex-fluidic device (P-VFD) has been developed to overcome the diffusion-limit and achieve fast reaction rates. The platform consists of a plasma oxazoline-coated polyvinyl chloride (POx-PVC) film with tough hydrogel covalently bound to its surface and a reactor tube for reactions. The hydrogel arrays on the film provide a strong matrix for entrapping biomolecules and tolerate shear stress, resulting in a >6-fold increase in reaction rate.

Dynamic Changes in Heparan Sulfate Nanostructure in Human Pluripotent Stem Cell Differentiation

Summary: Using expansion microscopy, researchers found that the nanotopography of heparan sulfate (HS) changes significantly during human pluripotent stem cell differentiation, and these changes are correlated with growth factor signaling. In undifferentiated cells, HS is densely distributed as hair-like protrusions, which can recruit fibroblast growth factor for signaling. However, during differentiation into ectodermal lineage, HS becomes localized into dispersed puncta, leading to a decrease in fibroblast growth factor binding to neural cells. This study highlights the importance of HS nanotopography in growth factor binding.

Magnetic Fe3O4@ZIF-8 nanoparticles as a drug release vehicle: pH-sensitive release of norfloxacin and its antibacterial activity

Summary: Metal-organic frameworks (MOFs) with high surface area and biodegradability have potential in drug release. Fe3O4 nanoparticles and MOFs composite materials, with magnetic features, were investigated for norfloxacin loading and release. The composite showed pH-sensitive properties and high release efficiency, making it suitable for targeted drug delivery. Its antibacterial activity against Escherichia coli was also tested, indicating its potential as a microbial inhibitor.

Sex differences in hippocampal structural plasticity and glycosaminoglycan disaccharide levels after neonatal handling

Absolute pharmacokinetics of heparin in primates

Summary: In this study, liquid chromatography-mass spectrometry (LC-MS) and the multiple reaction monitoring (MRM) method were used to directly measure heparin's concentration in non-human primates after administering porcine, bovine, and ovine heparin. The results showed that LC-MS and Heparin Red assay measurements closely correlated with unfractionated heparin's biological activities. This study provides a new method for measuring heparin concentration in plasma, which could improve our understanding of heparin's metabolism and dosing safety.

Summary: A ligand-mediated, spatially-confined synthesis method is developed to produce high-quality carbon dots from crude bioresources. The choice of solvent plays a critical role in the formation and properties of the carbon dots. The ligand-carbon interactions and electron transfer result in the formation of uniform colloidal carbon dots with multiple emissions. These carbon dots can be easily dispersed in various solvents and fabricated into thin-film devices, making them suitable for optoelectronic applications.

Anti-skin glycation and collagen level stimulation of brown seaweed extracts and their compositional characteristics

Summary: This study investigated the anti-skin aging potential of three Southern Australian brown seaweed extracts. The results showed that the extracts from Cystophora moniliformis and Cystophora siliquosa exhibited strong inhibitory effects on glycation activity and stimulated collagen production, suggesting their potential as skin anti-aging supplements. Further in vivo studies are warranted.
