Only a small fraction of the antibodies in a traditional polyclonal antibody mixture recognize the target of interest, frequently resulting in undesirable polyreactivity. Here, we show that high-quality recombinant polyclonals, in which hundreds of different antibodies are all directed toward a target of interest, can be easily generated in vitro by combining phage and yeast display. We show that, unlike traditional polyclonals, which are limited resources, recombinant polyclonal antibodies can be amplified over one hundred million-fold without losing representation or functionality. Our protocol was tested on 9 different targets to demonstrate how the strategy allows the selective amplification of antibodies directed toward desirable target specific epitopes, such as those found in one protein but not a closely related one, and the elimination of antibodies recognizing common epitopes, without significant loss of diversity. These recombinant renewable polyclonal antibodies are usable in different assays, and can be generated in high throughput. This approach could potentially be used to develop highly specific recombinant renewable antibodies against all human gene products.

A single donor is sufficient to produce a highly functional in vitro antibody library

M. Frank Erasmus, Sara D'Angelo, Fortunato Ferrara, Leslie Naranjo, Andre A. Teixeira, Rebecca Buonpane, Shaun M. Stewart, Horacio G. Nastri, Andrew R. M. Bradbury

Summary: The diversity of antibody complementarity determining regions is crucial for selecting high affinity leads, and increasing the number of donors is not the only way to enhance library diversity. This study demonstrates that lymphocyte numbers from a single healthy individual can be used to construct a diverse and functional antibody library.

A pandemic-enabled comparison of discovery platforms demonstrates a nai''ve antibody library can match the best immune-sourced antibodies

Summary: This study demonstrates that highly potent human neutralizing antibodies against the SARS-CoV-2 spike protein can be generated directly from a semisynthetic naive antibody library. These antibodies have comparable affinities, developability properties, and neutralization activities to those from immune sources, providing a potential alternative to traditional immunization methods.

Summary: The Omicron variant of SARS-CoV-2 has multiple mutations in the Spike protein, leading to increased transmissibility and evasion of antibodies. This study shows that vaccinated individuals who are infected with Omicron have robust immune responses, including both cellular and humoral immunity, as well as a new T cell response to non-Spike antigens.

Summary: Antibodies play a crucial role in research, diagnostics, and therapeutics. A high throughput method based on yeast display is described to select antibodies suitable for conversion to fluorescent chimeras. This method allows for the direct identification of functional antibodies in the single chain TGP format, which exhibit higher protein expression, easier purification, and one-step binding assays.

Drug-like antibodies with high affinity, diversity and developability directly from next-generation antibody libraries

Andre Azevedo Reis Teixeira, Michael Frank Erasmus, Sara D'Angelo, Leslie Naranjo, Fortunato Ferrara, Camila Leal-Lopes, Oliver Durrant, Cecile Galmiche, Aleardo Morelli, Anthony Scott-Tucker, Andrew Raymon Morton Bradbury

Summary: This paper introduces a new antibody library format that generates high-affinity binders with drug-like developability properties directly from initial selections, reducing the need for further engineering or affinity maturation. This innovative design is expected to accelerate drug development processes by reducing the failure rate of leads due to poor antibody affinities and developability.
