A proteomic study of peripheral blood mononuclear cells in systemic lupus erythematosus

Our objective was to analyze the changes in the protein expression profiles of peripheral blood mononuclear cells (PBMCs) in patients with systemic lupus erythematosus (SLE). Peripheral blood was obtained from patients with SLE and healthy controls. 2-D gel electrophoresis was performed, and gels were silver-stained. Differentially expressed protein spots were detected, some of which were identified by MALDI-TOF spectrometry. Match rates of 71% +/- 4% and 72% +/- 4% were gotten for control and patient gels, respectively. 791 17 spots were detected for control gels and 781 17 for patient gels. Eleven protein spots were Lip-regulated, and 9 protein spots were down-regulated in patients with SLE. Five differentially expressed proteins were identified as immunoglobulin J chain, apolipoprotein A-IV precursor, calprotectin L I H and zinc Finger protein Subfamily 1A (all up-regulated) and glutathione S-transferase (down-regulated), some of which had previously been shown to play a potential role in the pathogenesis of SLE. We conclude there are significant changes in the 2-D maps of PBMCs in patients with SLE and applying this proteomic approach may be a useful way to gain novel insights into SLE.

Summary: This study found a positive correlation between the expression of neurotransmitter receptors and relapse-free survival rates in breast cancer patients. Specifically, the expression of HTR6 was significantly decreased in advanced breast cancer. Further analysis revealed the potential regulatory role of HTR6 in signaling pathways and its association with the immune microenvironment. The study also identified possible reasons for the down-regulation of HTR6 in breast cancer.

Single-Cell RNA and ATAC Sequencing Reveal Hemodialysis-Related Immune Dysregulation of Circulating Immune Cell Subpopulations

Summary: This study integrated single-cell RNA sequencing (scRNA-seq) and single-cell assay for transposase-accessible chromatin sequencing (scATAC-seq) to investigate the transcriptional program and chromosomal accessibility of peripheral mononuclear cells (PBMCs) in maintenance hemodialysis patients. The study found that hemodialysis suppressed the expression of immune-related genes in CD4+ T cells and monocytes, inhibited downstream pathways of T cell receptor signaling, and altered cellular communication patterns between PBMC subgroups. Additionally, hemodialysis affected the chromatin accessibility profile and expression of transcription factors. This study provides a valuable framework for studying hemodialysis-related immune dysregulation and identifies potential therapeutic targets.

Exosomal circSHKBP1 participates in non-small cell lung cancer progression through PKM2-mediated glycolysis

Analyzing the gene regulatory network in hepatitis B patients by single-cell ATAC sequencing

The regulatory network of the hepatitis B signal pathway composed of genes linked to the differentially expressed peaks of these two CHB disease-specific cell types was drawn.

Differential expression of exosomal miRNAs and proteins in the plasma of systemic lupus erythematous patients

Summary: This study examined the expression profiles of exosomal miRNAs and proteins in the plasma of SLE patients and healthy subjects and discussed the underlying signaling network of circulating exosomes. The study identified differentially expressed exosomal miRNAs and proteins in SLE patients and healthy controls, as well as consensus molecules and signaling pathways. These findings indicate the potential application of exosomes in the clinical treatment and diagnosis of SLE disease.

Summary: This study analyzed and compared the spatial distribution of metabolites in healthy control and hepatitis B cirrhosis tissue samples using LC-MS/MS and mass spectrometry imaging system. The results showed that hepatitis B cirrhosis induced region-specific autophagy and dysregulation of unsaturated fatty acids, amino acids, and phosphatidylcholines metabolism. The mass spectrometry imaging system provided additional metabolites' spatial information, which can promote biomarker screening technology and support the exploration of novel mechanisms in liver cirrhosis.

Urinary microbiota and serum metabolite analysis in patients with diabetic kidney disease

Summary: By analyzing the microbiota and serum metabolites in urine samples from patients with diabetic kidney disease (DKD), it was found that DKD patients had a different urinary microbiota compared to the healthy controls, with the most abundant microbiota being Proteobacteria and Acidobacteria. Short-chain fatty acids and protein-bound uremic toxins were shown to be specific to DKD. Furthermore, the study revealed that the metabolism of arginine and proline may play a major role in the regulation of DKD.
