Leukotriene B4 is essential for selective eosinophil recruitment following allergen challenge of CD4+ cells in a model of chronic eosinophilic inflammation

Subcutaneous heat-coagulated egg white implants (EWI) induce chronic, intense local eosinophilia in mice, followed by asthma-like responses to airway ovalbumin challenge. Our goal was to define the mechanisms of selective eosinophil accumulation in the EWI model. EWI carriers were challenged i.p. with ovalbumin and the contributions of cellular immunity and inflammatory mediators to the resulting leukocyte accumulation were defined through cell transfer and pharmacological inhibition protocols. Eosinophil recruitment required Major Histocompatibility Complex Class It expression, and was abolished by the leukotriene B4 (LTB4) receptor antagonist CP 105.696, the 5-lipoxygenase inhibitor BWA4C and the 5-lipoxygenase activating protein inhibitor MK886. Eosinophil recruitment in EWI carriers followed transfer of: a) CD4(+) (but not CD4(-)) cells, harvested from EWI donors and restimulated ex vivo; b) their cell-free supernatants, containing LTB4. Restimulation in the presence of MK886 was ineffective. CC chemokine receptor ligand (CCL)5 and CCL2 were induced by ovalbumin challenge in vivo. mRNA for CCL17 and CCL11 was induced in ovalbumin-restimulated CD4(+) cells ex vivo. MK886 blocked induction of CCL17 Pretreatment of EWI carriers with MK886 eliminated the effectiveness of exogenously administered CCL11, CCL2 and CCL5. In conclusion, chemokine-producing, ovalburnin-restimulated CD4(+) cells initiate eosinophil recruitment which is strictly dependent on LTB4 production. (C) 2008 Elsevier Inc. All rights reserved.

Summary: Social isolation stress may facilitate oral squamous cell carcinoma (OSCC) occurrence and tumor progression, accompanied by increased levels of inflammatory mediators. The study found that isolated rats had higher OSCC occurrence rates, tumor volumes, and levels of inflammatory mediators than grouped rats. The levels of TNF-alpha, IL-6, IL1-beta, and MCP-1 in tumors were positively correlated with OSCC growth.

Summary: The study revealed the significant role of mast cells in periodontal disease-induced alveolar bone loss, especially in spontaneously hypertensive rats. Depletion of mast cells effectively inhibited alveolar bone loss caused by periodontal disease, reducing inflammatory status and mediator production, and decreasing osteoclast-related response markers.

Summary: This study found that the stress hormone NE induces resistance of oral cancer cells to cisplatin by acting on the ADRB/Akt/ABCG2 pathway. NE reduces the effectiveness of cisplatin on certain cell lines and achieves this resistance by pumping the drug out of the cell and inhibiting apoptosis.

Summary: This in vitro study evaluated the cytotoxicity potential of photodynamic therapy (PDT) using curcumin as a photosensitizer (PS) on fibroblasts. The results showed that curcumin PS at a concentration of 1000 mg/L was cytotoxic on L-929 fibroblast cells, while curcumin PS at a concentration of 500 mg/L had no influence on cell viability under in vitro conditions.

Summary: This study aimed to identify risk factors for mortality from SARS-CoV-2 infection, evaluate gene expression patterns of immune response pathways in COVID-19 patients, and identify circulating SARS-CoV-2 variants in a medium-sized city in Northeast Brazil. The study found that pre-existing comorbidity, male gender, cardiovascular disease, diabetes, chronic obstructive pulmonary disease, and PCR cycle threshold values under 22 were identified as risk factors for mortality. Furthermore, the study demonstrated that the severity of infection was associated with increased activation of inflammatory pathways, and multiple lineages of the virus were circulating.

Summary: This study evaluated the effects of melatonin (MEL) on the expression of TLR4, MyD88, TRIF, IRF-3, NF-kappa B, IL-1 beta and LPS, as well as the lipid profile in rats with apical periodontitis (AP) fed on a high-fat diet (HFD). MEL treatment reduced IRF-3 protein expression in the gastrocnemius muscle (GM) and IL-1 beta plasma concentration in the APMEL and HFDMEL groups. LPS plasma concentration decreased only in the HFDMEL group. Additionally, LDL decreased and HDL increased in the HFDMEL and HFDAPMEL groups. Treatment with MEL exhibited anti-inflammatory and anti-hyperlipidemic effects.

Summary: This study evaluated the antimicrobial action of losartan against C. albicans biofilms. The results showed that losartan inhibited the growth of C. albicans biofilms and had no significant toxicity on human cells. This indicates that losartan can be repurposed to interfere with the metabolism and development of Candida biofilms.
