The Aiolos transcription factor plays a crucial role in the control of lymphocyte differentiation and proliferation. The expression of Aiolos isoform has been studied in lymphoid pathologies but nothing is known about its expression in unaffected human hematopoietic subpopulations. In this manuscript we show for the first time the differential Aiolos expression at the RNA and protein level in hematopoietic cell subpopulations. B cells express higher levels of Aiolos than NK and T cells while monocytes express almost undetectable levels of Aiolos. Moreover, human CD34 (+) progenitors do not express Aiolos. We did not observe significant difference when comparing naive to memory T and B cells, but we observed an important difference between Bright and Dim NK cells. Furthermore, we show that, in addition to hematopoietic cells, non hematopoietic cell lines such as MCF-7, SW480, HEK, PC3 and HeLa also express Aiolos. (C) 2009 Elsevier Ltd. All rights reserved.

Summary: The study found that humoral immune responses to SARS-CoV-2 infection are dominated by IgA antibodies, with specific IgA plasmablasts rapidly expanding shortly after symptom onset and peaking in the third week of the disease. Virus-specific antibody responses include IgG, IgM, and IgA, with IgA playing a greater role in virus neutralization compared to IgG.

Summary: Our study found in autoimmune T1D models a consistent decrease in IL-17A, IL-22, and IL-23A in gut mucosa, along with impaired intestinal epithelial cell function and compromised gut integrity. This was accompanied by dysbiosis, including progressive loss of SFB. Anti-inflammatory treatment restored gut mucosa and immune cell function, reducing diabetes incidence.

Summary: Anaphylactic reactions to the novel mRNA lipid nanoparticle vaccines have raised significant public concern due to their unexpected occurrence, particularly among atopic individuals. Various potential contributors to these reactions have been considered, yet the clinical utility of available testing methods remains unclear. Further research and clinical trials are needed to address these concerns and ensure safety during public health emergencies.

Summary: The study suggests that there is a disruption in gut and systemic microbiota/immune balance in MS patients, with reduced IgA responses and increased IgG responses to gut bacteria. This breakdown of tolerance to gut microbiota may lead to enhanced systemic IgG immunity against commensals early in MS.
