Gene expression profiles of CD34+ cells were compared between 51 MDS patients and 7 controls. The most up-regulated genes in patients included HBG2, HBG1, CYBRD1, HSPA1B, ANGPT, and MYC, while 13 genes related to B-lymphopoiesis showed down-regulation. We observed in advanced MDS patients decreased expression of genes involved in cell cycle control, DNA repair and increased expression of proto-oncogenes, angiogenic and anti-apoptic genes. The results suggest that increased cell proliferation and resistance to apoptosis together with a loss of cell cycle control, damaged DNA repair and altered immune response may play an important role in malignant clone expansion in MDS. Microbiome analysis and predicted relative metabolomic turnover suggest bacterial heme and selenium metabolism are altered in the gastrointestinal system of zebrafish (Danio rerio) exposed to the organochlorine dieldrin

Qing Hua, Ondrej Adamovsky, Hana Vespalcova, Jonna Boyda, Jordan T. Schmidt, Marianne Kozuch, Serena L. M. Craft, Pamela E. Ginn, Stanislav Smatana, Eva Budinska, Maria Persico, Joseph H. Bisesi, Christopher J. Martyniuk

Summary: The study showed that environmentally-relevant doses of dieldrin did not significantly affect the gastrointestinal morphology and body weight of zebrafish, but altered the microbiome composition, including reductions in Firmicutes, Clostridia, and Betaproteobacteria. The predicted metabolic turnover analysis indicated potential impacts on bacterial heme biosynthesis and selenium metabolism pathways in the fish gastrointestinal system. Further research is needed to fully understand the long-term consequences of dietary exposure to dieldrin on microbiota composition.

Summary: National screening programs use dried blood samples to detect metabolic disorders or aberrant protein functions that may not be clinically evident in newborns, with a focus on gut microbiota metabolites and immunological acute-phase proteins. The delivery method (vaginal or cesarean section) plays a significant role in shaping microbial colonization and influencing the immune system's response and mucosal homeostasis in neonates. The study identified microbial xenobiotic receptors ligands in neonatal blood samples on the second day of life, indicating the potential importance of the human gut microbiota in maintaining immune system homeostasis.

Colorectal Tumour Mucosa Microbiome Is Enriched in Oral Pathogens and Defines Three Subtypes That Correlate with Markers of Tumour Progression

Summary: This study suggests that dysbiosis of the gut microbiome plays a role in the heterogeneity of colorectal cancer, affecting its phenotypic, prognostic, and treatment response aspects. The dominance of oral pathogenic bacteria in tumour mucosa of CRC patients was observed, while changes in stool microbiome were associated with clinical parameters such as lymph node involvement and synchronous metastases. The study identified new associations between microorganisms and CRC, providing insights for potential new treatments and biomarkers.

Summary: There are differences in microbiome composition and function related to metabolic health between individuals adhering to a vegan vs. omnivore diet. Vegans tend to have better glucose and lipid homeostasis profiles. The metabolomes of vegans and omnivores show significant differences, with vegans having lower abundance of potentially harmful metabolites and higher occurrence of potentially beneficial ones.

Summary: The study investigates the differences in microbial composition and diversity between male and female zebrafish. No significant differences were found in gut microbial diversity and predicted functional composition between the two sexes. However, there were differences in the abundance of certain bacterial families and genera. Transcriptome analysis revealed unique differentially expressed transcripts in the gastrointestinal tissue and differences in gene networks related to immune function and lipid metabolism between males and females.

RUNX1 mutations contribute to the progression of MDS due to disruption of antitumor cellular defense: a study on patients with lower-risk MDS

Summary: This study found that the RUNX1 mutation is the main predictor of rapid progression in LR-MDS patients. Comparison of gene expression profiles between patients with and without RUNX1 mutations suggests that RUNX1 mutations may trigger malignant transformation. Dysregulation of DDR and cellular senescence was also observed. Additionally, the expression profiles of RUNX1-mutated LR-MDS resemble those of higher-risk MDS.

Summary: This study comprehensively evaluated stool microRNA (miRNA) profiles as biomarkers for noninvasive colorectal cancer (CRC) diagnosis. A 5-miRNA signature accurately distinguished patients from control individuals and showed good correlation with tissue miRNA profiles and fecal profiles of different gastrointestinal diseases. These findings have the potential to improve noninvasive diagnosis and screening strategies for CRC.

Enrichment of human nasopharyngeal bacteriome with bacteria from dust after short-term exposure to indoor environment: a pilot study

Summary: This pilot study aimed to investigate the changes in human nasopharyngeal bacteriome after short-term exposure to indoor environments. Nasopharyngeal swabs and indoor dust samples were collected from 22 participants, and 16S rRNA sequencing analysis showed that short-term exposure to indoor environments can result in the enrichment of the nasopharynx with bacterial DNA from indoor dust. However, the bacterial composition varies by the indoor workplace environment.
