Hematopoiesis-related microRNA expression in myelodysplastic syndromes

MicroRNAs (miRNAs) are negative regulators of expression of genes involved in hematopoiesis. The present study sought to link hematopoiesis-relevant miRNAs with myelodysplastic syndromes (MDS) and MDS progression to acute myeloid leukemia (AML). We assessed 25 mature miRNAs in total RNA from bone marrow ( BM) and peripheral blood (PB) of 25 newly diagnosed patients with MDS and 12 controls. Twelve miRNAs in BM and six in PB were differentially expressed between patients with MDS and controls. Three of these miRNAs, belonging to the cluster 17-92, were overexpressed in both BM and PB. miR-15a in BM (p = 0.034) and miR-16 in PB (p = 0.005) were differentially expressed between low-risk and high-risk groups. miR-222 (p = 0.0023) and miR-181a (p = 0.014) expression was higher in AML than in MDS in both BM and PB. This study adds further evidence to the role of miRNAs in the pathogenesis of MDS and their transformation into AML.

This study provides the possibility of early detection for DLBCL patients at high risk of treatment failure.

The Undiagnosed Rare Disease Program of Catalonia project reanalyzed genomic data from 323 families with a neurologic rare disease, leading to a diagnosis for 20.7% of the patients.

Summary: This article presents the genomic characterization of 9 widely used human MPNST cell lines, providing valuable resources for translational research. NF1-related cell lines exhibited characteristics similar to primary MPNSTs, while sporadic cell lines showed different inactivation patterns of tumor suppressor genes and mutations. The re-classification of cell lines as melanomas and other sarcomas demonstrated different responses to drug treatment. Deep genomic analysis, methylome-based classification, and cell-identity marker expression challenged the identity of common MPNST cell lines, highlighting the need for revisions in MPNST diagnosis.

Generation of human iPSC-derived neurofibromaspheres for in vitro and in vivo uses

Summary: This study generated neurofibromaspheres by differentiating NF1(-/-) Schwann cells from induced pluripotent stem cells and combining them with neurofibroma primary fibroblasts. Neurofibroma-like tumors were developed when these neurofibromaspheres were engrafted in the sciatic nerve of nude mice. This model provides a versatile platform for drug screening and the study of neurofibroma biology.
