Summary: Controlling the internal structures of single-chain nanoparticles (SCNPs) is crucial for their design and synthesis. This study focuses on SCNPs containing near-infrared fluorescent dyes embedded in hydrophobic compartments. The formation of compartments is controlled by single-chain collapse and crosslinking, resulting in SCNPs with adjustable label densities. The specific conditions for single-chain collapse have a significant impact on the formation of desired core-shell structures and the internal compartments.

Summary: The properties and functions of non-covalent interaction-driven fluorescent supramolecular self-assembly depend greatly on their evolution dynamics. Electron microscopy, atomic force microscopy, and confocal laser scanning microscopy have been used to elucidate the formation of molecular self-assembly. However, some pertinent issues often impede the real-time analysis and exploration of the dynamics of molecular self-assembly processes. Fluorescence correlation spectroscopy and fluorescence lifetime imaging microscopy have recently been explored to unravel the physical picture of the in situ growth dynamics and stimuli-induced morphological transformation of luminescent self-assembled structures.

Summary: The study demonstrates the existence of nanocompartments within SCNPs, achieved by encapsulating labels within water-dispersible SCNPs using amphiphilic polymers and copper-catalyzed synthesis. A detailed profile of the interior of the SCNPs and the labels' microenvironment was obtained through EPR experiments.

Summary: Typical studies of gastral toxicity of nanoparticles are expensive and difficult. Fluorescence labeling can be an alternative, but most fluorophores are sensitive to changes in pH and hydrolysis reactions in mammalian digestive tracts. A new pH insensitive and hydrolitically stable fluorophore, mDTEB, based on BODIPY center, has been synthesized and validated for toxicity studies.

Summary: RNA plays a crucial role in cellular processes, making it essential to visualize and quantify its molecular dynamics directly in living cells to understand its metabolism. In this study, fluorescence correlation methods were utilized to demonstrate the visualization and quantification of interactions between ECHO probes targeting mRNA, showing changes in fluorescence brightness and diffusion coefficient upon binding. The uptake of ECHO probes in HeLa cells for mRNA detection provides new insights into intracellular mRNA metabolism.
